KARAKTER FENOTIPIK DAN VARIASI GENETIK MELON (Cucumis melo L. kultivar GMP) HASIL INDUKSI BIOCATHARANTINE BERDASARKAN INTER-SIMPLE SEQUENCE REPEAT

Kultivar Gama Melon Parfum (GMP) merupakan kultivar hasil persilangan antara melon Natsu no Omoide NO3 betina dan Miyamauri MR5 jantan. Karakteristik yang dimiliki oleh melon kultivar GMP adalah aroma buah yang sangat harum seperti parfum. Fakultas Biologi UGM telah bekerjasama dengan PT. Gizi Indonesia pada tahun 2019 untuk mengembangkan melon kultivar GMP sebagai bahan baku industri kosmetik. Inovasi pengembangan ekstrak buah melon kultivar GMP sebagai raw material produk kosmetik organik tersebut melahirkan ide dalam pemuliaan tanaman, yaitu induksi Biocatharantine sebagai agen antimitotik dengan harapan akan memicu terjadinya poliploidisasi sehingga terjadi peningkatan kuantitatif maupun kualitatif pada tanaman, khususnya pada buah yang dihasilkan. Penelitian ini berlokasi di Madurejo, Prambanan, Yogyakarta dan Laboratorium Genetika dan Pemuliaan Fakultas Biologi UGM. Metode yang digunakan dalam penelitian ini adalah perendaman benih dalam larutan Biocatharantine dengan konsentrasi 0%; 0,05%; 0,1%; dan 0,5% selama 24 jam. Analisis karakter fenotipik dilakukan dengan oneway ANOVA dan dilanjutkan dengan uji Duncan pada taraf signifikansi 5%. Karakterisasi molekuler dilakukan melalui metode PCR-ISSR selanjutnya dianalisis dengan program MVSP (Multivariate Statistical Package). Hasil Konsentrasi paling efektif poliploidisasi dengan induksi Biocatharantine adalah pada konsentrasi 0,1% yang menghasilkan peningkatan berat segar buah, yaitu 224,00 gram. Analisis kekerabatan fenetik menyatakan bahwa melon kultivar GMP perlakuan kontrol dan induksi Biocatharantine memiliki similaritas 82,1%.

Gama Melon Parfum (GMP) are cultivars resulted from crossing between melons Natsu no Omoide NO3 female and Miyamauri MR5 male. The characteristic possessed by the GMP melon cultivar is the smell of a very fragrant fruit like perfume. The Faculty of Biology UGM has collaborated with PT. Gizi Indonesia since 2019 to develop melon cultivar GMP as a raw material for cosmetic industry. The innovation in the development of 'GMP' fruit extract as a raw material for organic cosmetic products gave birth to the idea of plant breeding, namely the induction of Biocatharantine as an antimatotic agent in the hope that it would trigger polyploidization so that there are increases in quantitative and qualitative parameters, especially in the fruits produced. This research is located in Madurejo, Prambanan, Yogyakarta and the Genetics and Breeding Laboratory of the Faculty of Biology UGM. The method used in this study was soaking melon cultivar GMP seeds in Biocatharantine solution with a concentration of 0%; 0.05%; 0.1%; and 0.5% for 24 hours. Phenotypic character analysis was performed with oneway ANOVA and continued with duncan test at significance level of 5%. Molecular characterization was carried out through PCR-ISSR method and then analyzed with the MVSP (Multivariate Statistical Package) program. The most effective concentration of polyploidization with Biocatharantine induction was at concentration of 0.1% which resulted in an increase in the fresh weight of the fruit, which was 224.00 grams. Phenetic relationship analysis stated that melon 'GMP' control and induction of Biocatharantine had a similarity of 82.1%.
