Efektivitas Ekstrak Kokon Attacus atlas Linnaeus, 1758 sebagai UV Protektan Bacillus thuringiensis Serotipe kurstaki Pengendali Spodoptera litura (Fabricius, 1775) di Bawah Paparan Sinar Matahari

Bacillus thuringiensis merupakan salah satu bakteri entomopatogen yang umumnya digunakan untuk mengontrol serangga hama dan serangga vector, salah satunya adalah ulat grayak, S. litura (Fab.). Namun, keberadaan B. thuringiensis mudah terdegradasi oleh sinar UV, terlebih jika terpapar di bawah sinar matahari secara langsung, sehingga B. thuringiensis memerlukan perlindungan sebagai UV protektan. Tujuan dari penelitian ini adalah untuk mempelajari keefektifan penambahan ekstrak serisin yang berasal dari kokon Attacus atlas terhadap mortalitas S. litura, pengamatan subletal pada larva S. litura dan persistensi keberadaan B. thuringiensis. Penelitian ini menggunakan formulasi 2,5% ekstrak kokon A. atlas dengan B. thuringiensis serotipe kurstaki strain HD-7 yang didapatkan dari produk komersial DiPel-WP® (Abbot Co., IN). Formulasi tersebut dipapar di bawah sinar matahari secara langsung selama 0, 1, 2, 3 minggu. Pengujian dilakukan dengan menggunakan 20 individu larva S. litura instar satu pada setiap ulangan. Setiap perlakuan masing masing terdiri dari 5 ulangan. Scanning Electron Microscope (SEM) dilakukan untuk mengamati persistensi B. thuringiensis dan keberadaan ekstrak kokon A. atlas. Kematian S. litura dihitung pada hari ke-3, dan pengamatan subletal diamati pada hari ke-7 hingga menjadi fase pupa dan imago. Adapun metode pengamatan SEM dimulai dari sampel yang divakum, sampel dilapisi dengan Au, dan pengamatan pada layar komputer. Hasil uji patogenisitas B. thuringiensis terhadap larva instar satu S. litura menyebutkan bahwa penambahan ekstrak pada perlakuan dipapar dan tidak dipapar menunjukkan perbedaan nyata pada α=0,05. Hasil analisis korelasi menunjukkan semakin lama paparan ekstrak maka akan menurunkan mortalitas S. litura. Mortalitas S. litura dengan penambahan ekstrak pada B. thuringiensis selama waktu paparan tiga minggu mencapai 39,39%. Efek subletal yang ditemukan pada S. litura akibat paparan B. thuringiensis adalah penurunan nafsu makan, bentuk pupa yang cacat serta sayap imago keriting. Keberadaan B. thuringiensis yang diamati menggunakan mikroskop fase kontras menunjukkan persistensi nya hingga paparan minggu ke-3. Pengamatan SEM menunjukkan semakin lama paparan, maka B. thuringiensis semakin terdegradasi dan permukaannya terlihat lebih halus. Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak kokon efektif sebagai UV protektan B. thuringiensis hingga waktu paparan 3 minggu.

Bacillus thuringiensis serotype kurstaki is an entomopathogenic bacteria that commonly used to control pests and vector insects, one of them is the army worm Spodoptera litura (Fab.). However, B. thuringiensis might be easily degraded by exposed sunlight. The aims of this research were to determine the effectiveness of A. atlas L. cocoon extract as UV protectant B. thuringiensis on the pathogenicity of S. litura, observed the sub-lethal effect of S. litura, and observed the persistence of B. thuringiensis. This research was using a preparation from 2.5% of A. atlas cocoon extract and B. thuringiensis serotype kurstaki strain HD-7 formulation obtained from commercial product DiPel-WP® (Abbot Co., IN). The Bt preparations were exposed to sunlight for 0, 1, 2, or 3 weeks. The pathogenicity test was conducted against 20 individuals of first larvae instar of S. litura per replicate using five replicates in the artificial diet. The mortality of S. litura were counted on the third day after insecticide application and the sub-lethal effect of S. litura was observed from the seventh day of application until the insects pupate and developing into imago. The scanning electron microscope (SEM) analysis began with vacuuming the sample, then sample coating and finally observing the samples on the SEM. The result showed that the pathogenicity test of B. thuringiensis S. litura were significant difference (ANOVA, α<0.05) between UV exposed and without UV exposed treatments. The correlation analysis showed that increasing time exposure will decreased mortality of S. litura. The mortality of S. litura treated with 2.5% A. atlas cocoon extract added to B. thuringiensis formulation was 39.39%. The sub-lethal effect showed decreasing appetite, pupal deformation and emerging of curly wings imago of S. litura. B. thuringiensis persistence during three weeks of exposure. Furthermore, SEM analysis showed that correlated to the length of UV exposure, B. thuringiensis preparations were more degraded as shown by a smoother particle surface of the preparations. Based on these data, it can be concluded that the addition of A. atlas cocoon extract is effective for UV protection during three weeks of exposure.
