Produktivitas Telur dan Deteksi Gen PRL pada Ayam F2 Hibrida Mahkota (Gallus gallus domesticus Linnaeus, 1758) Hasil Persilangan Resiprok

Keanekaragaman jenis ayam lokal di Indonesia mempunyai potensi pemanfaatan yang sangat luas. Permintaan terhadap produk peternakan baik daging dan telur semakin meningkat sehingga program pengembangan dan intensifikasi ayam lokal memiliki prospek yang baik. Ayam lokal memiliki kualitas daging dan telur yang lebih baik dibandingkan ayam ras. Produktivitas daging dan telur yang rendah menjadi hambatan dalam pengembangan ayam lokal. Ayam mahkota memiliki karakteristik telur menyerupai ayam lokal sehingga tujuan dilakukan penelitian ini adalah untuk mengetahui produktivitas telur ayam F2 Hibrida Mahkota dan melihat polimorfisme indel pada promoter PRL sebagai penanda sifat produktivitas telur serta melakukan analisis kualitas telur. Tahapan yang dilakukan pada penelitian ini yaitu persilangan ayam, pemeliharaan ayam, pengamatan produksi telur, analisis kualitas telur, uji proksimat, isolasi DNA, amplifikasi DNA dengan PCR, dan elektroforensis. Hasil penelitian menunjukan rerata kumulatif jumlah telur ayam F2 Hibrida Mahkota selama 8 minggu adalah 42,0 butir, lebih rendah dibandingkan F1 Hibrida Mahkota 48 butir dan lebih tinggi dibandingkan Pelung 27 butir. Kualitas telur F2 Hibrida Mahkota terdiri dari rerata berat telur 39,75±2,27g, egg shape index 60% berbentuk normal, berat dan ketebalan kerabang 3,72±0,31g dan 0,23±0,02mm. Kandungan nutrisi telur F2 Hibrida Mahkota terdiri dari air 73,25%, abu 1,55%, protein total 10,54%, lemak 9,77%, karbohidrat 4,90% dan kalori 141,3Kal/100g. Terdapat dua variasi polimorfisme indel pada promoter PRL yaitu delesi (DD) dan insersi-delesi (ID). Mutasi yang ditemukan pada sampel F2 Hibrida Mahkota adalah mutasi delesi.

The diversity of local chicken species in Indonesia has a very wide utilization potential. The demand for livestock products, both meat and eggs, is increasing so that the local chicken development and intensification program has good prospects. Local chickens have better meat and egg quality than purebred chickens. The low productivity of meat and eggs is an obstacle in the development of local chickens. Crested Chickens have egg characteristics resembling local chickens, so the purposed of this study was to determined the egg productivity of F2 Crested Chicken Hybrid and to see indel polymorphisms in the PRL as a marker of egg productivity characteristics and to analyze egg quality. The procedures carried out in this study were chicken crossing, chicken rearing, egg production observation, egg quality analysis, proximate test, DNA Isolation, DNA Amplification by PCR, and Electrophoresis. The results showed that the cumulative average of the number of eggs of F2 Crested Chicken Hybrid for 8 weeks was 42.0 eggs, lower than F1 Crested Hybrid 48 eggs and higher than Pelung 27 eggs. The quality of the eggs of F2 Crested Chicken Hybrid consisted of an average egg weight of 39.75±2.27g, an egg shape index of 60% normal shape, egg weight and thickness of 3.72±0.31g and 0.23±0.02mm. The nutritional content of the F2 Crested Hybrid egg consists of 73.25% water, 1.55% ash, 10.54% total protein, 9.77% fat, 4.90% carbohydrates and 141.3 calories. There are two indel polymorphism in PRL promoter are deletion (DD) and insertion-deletion (ID). The mutation found in the F2 Crested Chicken Hybrid is deletion mutation.
