Effects of Wickerhamomyces anomalus Inoculation in Coffee Fermentation on Microbiological Profile and Physicochemical Properties of Arabica Coffee Beans (Coffea arabica)

Fermentasi merupakan suatu proses yang penting untuk menghasilkan kopi berkualitas tinggi dan sangat dipengaruhi oleh aktivitas mikrobia. Salah satu langkah untuk meningkatkan proses fermentasi kopi adalah inokulasi kultur mikrobia. Wickerhamomyces anomalus merupakan kapang (yeast) lokal yang ada di dalam kopi Arabika dan memiliki aktivitas pektinolitik, penghambat pertumbuhan Aspergillus ochraceus, produksi komponen volatile, dan modifikasi prekursor flavor. Penelitian ini bertujuan untuk mengeksplorasi pengaruh inokulasi Wickerhamomyces anomalus terhadap profil mikrobiologi dan sifat fisikokimia biji kopi, terbatas pada pH, total asam, dan kandungan asam klorogenat. Sejumlah 108 CFU/ml kultur Wickerhamomyces anomalus ditambahkan ke dalam wadah fermentasi dengan komposisi 100g biji kopi dan 200 ml air. Fermentasi metode olah basah dilakukan selama 48 jam dan diambil sampel setiap 12 jam. Profil mikrobiologi dari fermentasi metode olah basah dengan inokulasi Wickerhamomyces anomalus mampu meningkatkan pertumbuhan yeast pada 12 jam, mencapai 9.74 log CFU/ml, dengan pH 4.08 dan total asam 1.97%. Analisis untuk pH, total asam, dan asam klorogenat dari biji kopi menunjukkan tidak adanya perbedaan signifikan pada sampel dengan inokulasi. Namun, sampel dengan inokulasi memiliki pH lebih rendah, total asam lebih tinggi, dan konsentrasi asam klorogenat lebih rendah daripada sampel control. Wickerhamomyces anomalus juga mampu membentuk komponen flavor seperti etil asetat dan menunjukkan aktivitas pektinolitik melalui pembentukan enzim poligalakturonase. Penelitian lebih lanjut untuk eksplorasi komponen volatil, perubahan komponen kimia yang berhubungan dengan flavor, dan enzim pektinolitik yang diproduksi oleh Wickerhamomyces anomalus dalam fermentasi kopi perlu dilakukan.

Fermentation is a crucial process to develop high quality coffee beans and is widely influenced by microbial activity. One of the approaches to improve coffee fermentation is culture inoculation. Wickerhamomyces anomalus is an indigenous yeast present in Arabica coffee beans that has shown pectinolytic activity, inhibitory effects on Aspergillus ochraceous, volatile compounds production, and flavor precursors modification. This research aims for exploring Wickerhamomyces anomalus effect on the microbiological and physicochemical properties of coffee beans, limited to pH, total acid, and chlorogenic acid concentration. Wickerhamomyces anomalus culture (108 CFU/ml) was added to the fermentation container with the composition of 100 g coffee beans and 200 ml water. Wet fermentation was conducted for 48 hours, and samples were taken every 12 hours. The microbiological profile of coffee fermentation with Wickerhamomyces anomalus inoculation promoted yeast population growth at 12 hours, reaching 9.74 log CFU/ml, with a pH value of 4.08 and a total acid value of 1.97%. The pH, total acid, and chlorogenic acid concentrations of coffee beans showed no significant difference for inoculated samples. However, inoculated coffee green beans showed lower pH value, higher total acid value, and lower chlorogenic acid concentration after 48 hours of fermentation, if compared to the control. Furthermore, Wickerhamomyces anomalus is also discovered to produce flavor compounds, including ethyl acetate, and display pectinolytic activity by polygalacturonase enzyme production. Further exploration of volatile compounds, changes of other chemical compounds related to flavor, and pectinolytic enzyme produced by Wickerhamomyces anomalus in coffee fermentation should be conducted.
