Profil Hematologi Koi (Cyprinus carpio Linnaeus, 1758)

Penelitian ini bertujuan untuk mengetahui profil hematologi Koi. Sampel yang digunakan adalah koi dari Gumulan Koi Farm, Kabupaten Klaten, Provinsi Jawa Tengah, SNS Koi Farm, Kabupaten Bantul, dan DIY Hatchery Koi, Kabupaten Sleman, Provinsi Daerah Istimewa Yogyakarta. Parameter yang diamati meliputi jumlah sel darah merah, jumlah sel darah putih, kadar hemoglobin, hematokrit, leukokrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin centration (MCHC), aktivitas fagositosis, indeks fagositosis, ledakan respirasi ekstraseluler, aktivitas superoksida dismutase, aktivitas lisozim, aglutinasi alami, aktivitas antibakteri serum, dan total protein plasma. Hasil menunjukkan jumlah sel darah merah 0,57 – 1,28 x 106 sel/mm3, jumlah sel darah putih 38,01 – 45,28 x 103 sel/mm3, hematokrit 22,10 – 36,31%, leukokrit 1,74 – 3,25%, kadar hemoglobin 60,50 – 93,50 g/l, MCV 286,58 – 525,23 fl, MCH 75,02 – 107,97 pg, MCHC 162,45 – 275,03 g/l, aktivitas fagositosis 10,25 – 13,00%, indeks fagositosis 1,09 – 1,14, ledakan respirasi ekstraseluler 0,188 – 0,263, aktivitas SOD 0,1375 – 0,1411, neutrofil 14,25 – 18,50%, monosit 3,25 – 4,50%, limfosit 77,75 – 81,25%, aktivitas lisozim 26,147 – 26,196 U/ml, aglutinasi alami 3,00 – 4,75, TPP 44,50 – 56,40 mg/ml, dan aktivitas antibakteri serum 36,84 – 41,45%. Jumlah sel darah merah, hematokrit, leukokrit, MCV, dan aglutinasi alami menunjukkan perbedaan yang signifikan (P<0,05) antar lokasi tetapi masih dalam batas normal. Jumlah sel darah putih, kadar hemoglobin, MCH, MCHC, aktivitas fagositosis, indeks fagositosis, persentase monosit, limfosit, dan neutrofil, ledakan respirasi ekstraseluler, aktivitas SOD, aktivitas lisozim, aktivitas antibakteri serum, dan total protein plasma tidak menunjukkan perbedaan yang signifikan (P>0,05). Koi dari ketiga lokasi dalam keadaan sehat.

The purpose of this study was to determine the hematological profile of koi. The samples used were koi from Gumulan Koi Farm, Klaten Regency, Jawa Tengah Province, SNS Koi Farm, Bantul Regency, and DIY Hatchery Koi, Sleman Regency, Daerah Istimewa Yogyakarta Province. Parameters observed were red blood cell count, white blood cell count, hemoglobin level, hematocrit, leukocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin centration (MCHC), phagocytic activity, phagocytic index, respiratosy burst, superoxide dismutase (SOD) activity, lysozyme activity, natural agglutination, serum antibacterial activity, and total plasma protein. The results showed the number of red blood cells 0.57 – 1.28 x 106 cells/mm3, the number of white blood cells 38.01 – 45.28 x 103 cells/mm3, hematocrit 22.10 – 36.31%, leukocrit 1.74 – 3 ,25%, hemoglobin level 60.50 – 93.50 g/l, MCV 286.58 – 525.23 fl, MCH 75.02 – 107.97 pg, MCHC 162.45 – 275.03 g/l, phagocytotic activity 10.25 – 13.00%, phagocytosis index 1.09 – 1.14, respiratory burst 0.188 – 0.263, SOD activity 0.1375 – 0.1411, neutrophils 14.25 – 18.50%, monocytes 3, 25 – 4.50%, lymphocytes 77.75 – 81.25%, lysozyme activity 26,147 – 26,196 U/ml, natural agglutination 3.00 – 4.75, TPP 44.50 – 56.40 mg/ml, and serum antibacterial activity 36.84 – 41.45%. Red blood cell count, hematocrit, leukocrit, MCV, and natural agglutination showed significant differences (P<0.05) between locations but were still within normal limits. White blood cell count, hemoglobin level, MCH, MCHC, phagocytic activity, phagocytosis index, percentage of monocytes, lymphocytes, and neutrophils, extracellular respiration burst, SOD activity, lysozyme activity, serum antibacterial activity, and total plasma protein did not show significant differences (P>0.05). Koi from all locations are in good health.
