Interaksi Populasi Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) dengan Gejala Penyakit Kuning pada Pertanaman Cabai (Capsicum frutescens L.) di Dataran Tinggi

Insidensi penyakit kuning pada tanaman cabai di Daerah Istimewa Yogyakarta dilaporkan mencapai 100%. Bemisia tabaci (Hemiptera: Aleyrodidae) merupakan vektor penyakit kuning. B. tabaci disebut sebagai cryptic species complex dan memiliki kekhasan tersendiri pada setiap daerah. Insidensi penyakit kuning diduga tidak selalu berkaitan dengan tingginya populasi serangga vektor. Penelitian ini bertujuan untuk mengetahui interaksi populasi B. tabaci dengan gejala penyakit kuning pada tanaman cabai dan melakukan identifikasi keragaman genetik B. tabaci di daerah dataran tinggi. Metode yang digunakan meliputi pengamtan insidensi dan intensitas penyakit kuning, penghitungan populasi B. tabaci per tanaman, pengumpulan sampel imago, ekstraksi DNA, identifikasi keragaman genetik B. tabaci dan deteksi Begomovirus dengan teknik PCR, analisis filogenetik gen mtCOI dan koefisien regresi. Hasil penelitian menunjukkan bahwa interaksi populasi B. tabaci dengan insidensi penyakit kuning pada umur tanaman cabai 20 hst menunjukkan hubungan yang kuat sedangkan pada umur tanaman cabai 50, 70, dan 85 hst menunjukkan keeratan hubungan yang rendah. Populasi B. tabaci saat perkembangan penyakit mulai tinggi, cenderung mengalami penurunan. Identifikasi secara cepat menggunakan primer spesifik biotipe menunjukkan adanya keragaman genetik B. tabaci di dataran tinggi Kaliurang yaitu biotipe Q yang masih menjadi dugaan awal. Namun, berdasarkan analisis fragmen mtCOI hanya ditemukan satu genotipe B. tabaci di dataran tinggi Kaliurang yaitu biotipe non B yang termasuk dalam haplotipe Asia I.

The incidence of yellow disease chili in Yogyakarta reported to reach 100%. Bemisia tabaci (Hemiptera: Aleyrodidae) is a vector of yellow disease and called a cryptic species complex and has its own peculiarities in each area. The incidence of yellow disease is thought to be not always related to the high population of vector insects. This study aims to determine the interaction of the B. tabaci population with symptoms of yellow disease in chili and identify the genetic diversity of B. tabaci in the highlands. The methods used include observing the yellow disease, calculating and collecting the population of vector, extracting DNA, identifying the genetic diversity of B. tabaci and detecting Begomovirus, and analysis of the mtCOI gene. The results showed that the interaction of population of B. tabaci with the incidence of yellow disease at the age of chili plants 20 days after planting showed a strong relationship, while at the age of chili plants 50, 70, and 85 days after planting, the correlation was low. The population of B. tabaci at the time of high disease progression tends to decrease. Rapid identification using biotype-specific primers showed biotypes Q in the highland, which are still preliminary estimates. However, based on the analysis of mtCOI fragments, only one genotype of B. tabaci was found in the Kaliurang highlands, namely the non B biotype or the Asia I haplotype.
