OPTIMASI PRODUKSI ANTIBIOTIK OLEH BAKTERI ENDOFIT TANAMAN BENGLE (Zingiber cassumunar) MENGGUNAKAN MEDIUM TETES TEBU DAN IDENTIFIKASI BAKTERINYA

Antibiotik adalah metabolit sekunder yang disintesis oleh mikroba yang dapat menghambat atau membunuh mikroba lain yang hidup di lingkungan. Bakteri endofit adalah bakteri yang hidup dalam jaringan tanaman tanpa menimbulkan penyakit pada inangnya dan berpotensi sebagai sumber antibiotik baru. Penelitian ini bertujuan untuk mengetahui kemampuan bakteri endofit hasil isolasi (penelitian sebelumnya) dalam pemanfaatan sumber karbon (C) dari tetes tebu dan pengaruh substrat terhadap produksi metabolit sekunder (antibiotik) pada tiga medium pertumbuhan yang berbeda. Optimasi terdiri dari konsentrasi inokulum, konsentrasi substrat, dan kondisi lingkungan (pH, suhu, dan agitasi). Metode penelitian dimulai dengan seleksi isolat berdasarkan aktivitas enzim invertase; produksi antibiotik pada medium Glucose Yeast (GY), Nutrient Broth (NB), dan Antibiotik-3; daya hambat antibiotik terhadap tiga macam bakteri indikator (Fusarium oxysporum, Bacillus subtilis, dan Candida albicans), serta identifikasi isolat terpilih berdasarkan analisis gen 16S rRNA. Hasil percobaan menunjukkan isolat terpilih adalah BBLEB dan dilanjutkan ketahap optimasi dengan konsentrasi inokulum 5%, konsentrasi substrat 3%, pH 8, suhu 30°C, dan agitasi 175 rpm dapat meningkatkan produksi antibiotik dengan daya hambat dari 3,0 menjadi diatas 4,0. Hasil identifikasi berdasarkan analisis gen 16S rRNA menunjukan bahwa isolat BBLEB teridentifikasi sebagai Pseudomonas aeruginosa.

Antibiotics are secondary metabolites synthesized by microbes that can inhibit or kill other microbes that live in the environment. Endophytic bacteria are bacteria that live in plant tissues without causing disease in the host and potential as a source of new antibiotics. This study was aimed to determine the ability of the isolated and selected endophytic bacteria (from the previous study) in the utilization of carbon source (C) from sugarcane molasses and the effect of the substrate on the production of secondary metabolites (antibiotics) in three different media. The optimization consists of inoculum concentration, substrate concentration, and environmental conditions (pH, temperature, and agitation). The method of the research were selection of isolates based on the invertase enzyme activity; antibiotic production on Glucose Yeast (GY), Nutrient Broth (NB), and Antibiotics-3; inhibition effect of antibiotics to three kinds of bacterial indicator that were (Fusarium oxysporum, Bacillus subtilis, and Candida albicans), and identification based on the 16S rRNA gene analysis. Experimental results showed that BBLEB isolate was chosen as selected strain, and furthermore optimization step by inoculum concentration 5%, substrate concentration 3%, pH 8, temperature 30°C, and agitation 175 rpm could increase the antibiotic production with inhibition effect of antibiotic from 3.0 to more than 4.0. The identification result based on the 16S rRNA gene analysis showed that BBLEB was identified as Pseudomonas aeruginosa.
