Identification of Antibacterial Peptide in the HPLC Fraction of Trypsin Hydrolysate Protein Produced from Acid Extraction of Jatropha Seeds

Telah dilakukan fraksinasi menggunakan High-performance Liquid Chromatography (HPLC) terhadap hidrolisat tripsin protein biji jarak kepyar hasil ekstraksi trifloroasetat. Urutan asam amino peptida pada fraksi dengan aktivitas antibakteri tinggi diidentifikasi sebagai kandidat antibiotik baru. Protein dari biji jarak diekstrak menggunakan TFA dan selanjutnya dihidrolisis dengan tripsin. Fraksinasi HPLC menggunakan sistem fase balik dengan fase gerak gradien menggunakan ACN 6% dalam TFA 0,1% dan ACN 60% dalam TFA 0,1%. Uji aktivitas antibakteri fraksi peptida dilakukan dengan metode mikrodilusi terhadap Escherichia coli dan Staphylococcus aureus. Peptida dari fraksi dengan aktivitas antibakteri tinggi diidentifikasi dengan Liquid Chromatography-High Resolution Mass Spectroscopy (LC-HRMS) dan analisis bioinformatik. Hasil penelitian menunjukkan bahwa ekstraksi TFA protein biji jarak kepyar menghasikan rendemen sebesar 61%, sedangkan hidrolisis dengan tripsin menghasilkan nilai derajat hidrolisis sampai dengan 65,9%. Berdasarkan puncak- puncak pada profil kromatogram fraksinasi hidrolisat protein menghasilkan 10 fraksi. Fraksi hidrolisat dengan aktivitas antibakteri tinggi ditunjukan oleh fraksi F5 dan F8 dengan nilai MIC masing-masing â�¤ 4,32 dan â�¤ 1,42 �¼g/mL. Urutan asam amino peptida yang yang diduga berpotensi sebagai peptida antimikroba adalah AVGHNWK, KPGVRKPPKR, AAGASG, dan APPPAK.

Fractionation has been carried out using High-performance Liquid Chromatography (HPLC) on trypsin hydrolysate of jatropha seed protein extracted with trifluoroacetate. The amino acid sequence of the peptide in the fraction with high antibacterial activity was identified as a new antibiotic candidate. Protein from jatropha bean was extracted using TFA and then hydrolyzed with trypsin. HPLC fractionation used a reverse phase system with gradient mobile phase consist of 6% ACN in 0.1% TFA and 60% ACN in 0.1% TFA. The antibacterial activity test of the peptide fraction was carried out by microdilution method against Escherichia coli and Staphylococcus aureus. Peptides from the fraction with high antibacterial activity were identified by Liquid Chromatography-High Resolution Mass Spectroscopy (LC-HRMS) and bioinformatic analysis. The results showed that TFA extraction of jatropha seed protein resulted in a yield of 61%, while hydrolysis with trypsin resulted in a degree of hydrolysis up to 65.9%. Based on the peaks in the chromatogram profile, the protein hydrolysate fractionation resulted in 10 fractions. The hydrolysate fraction with high antibacterial activity was indicated by the F5 and F8 fractions with MIC values �4.32 and �1.42 g/mL, respectively. The amino acid sequences of the peptides thought to be potential antimicrobial peptides were AVGHNWK, KPGVRKPPKR, AAGASG, and APPPAK.
