Penelitian ini bertujuan untuk mengetahui pengaruh penambahan ekstrak temulawak (Curcuma xanthorrhiza) dalam bahan pengencer sitrat kuning telur terhadap kualitas spermatozoa domba ekor tipis. Semen segar diperoleh dari domba ekor tipis berumur 3 tahun yang ditampung menggunakan vagina buatan. Semen segar yang diperoleh diencerkan menggunakan sitrat kuning telur dengan penambahan ekstrak temulawak. Perlakuan terdiri dari pengencer sitrat kuning telur sebagai kontrol (P0), kombinasi sitrat kuning telur dengan ekstrak temulawak 1% (P1), 2% (P2), dan 3% (P3). Semen yang telah diencerkan kemudian disimpan di refrigerator dengan suhu 5 derajat C. Variabel yang diamati yaitu kualitas spermatozoa domba seperti motilitas (%), viabilitas (%), dan abnormalitas (%). Persentase motilitas spermatozoa ditentukan secara subyektif kuantitatif. Pemeriksaan viabilitas dan abnormalitas spermatozoa menggunakan pewarnaan eosin. Kualitas spermatozoa dievaluasi setelah 3 jam dan 3 hari setelah penyimpanan. Data yang diperoleh dianalisis dengan analisis variansi menggunakan Rancangan Acak Lengkap (RAL) pola searah dan dilanjutkan dengan uji lanjut menggunakan Duncan Multiple Range Test. Hasil analisis menunjukkan bahwa penambahan ekstrak temulawak menunjukkan perbedaan yang nyata (p<0,05) terhadap motilitas, viabilitas, dan abnormalitas spermatozoa setelah penyimpanan pada suhu 5 derajat C. Kualitas spermatozoa yang diamati pada jam ke 3 menunjukkan bahwa penambahan ekstrak temulawak 3% memberikan hasil terbaik terhadap motilitas, viabilitas, dan abnormalitas spermatozoa berturut-turut yaitu 80,00+-2,00, 84,57+-1,10 dan 7,82+-0,68%. Penambahan ekstrak temulawak 3% juga memberikan hasil terbaik terhadap motilitas, viabilitas, dan abnormalitas spermatozoa yang diamati pada hari ke 3 berturut-turut yaitu 61,67+-2,88, 77,54+-0,98 dan 14,47+-0,90%. Penambahan 3% ekstrak temulawak memberikan kualitas semen terbaik setelah penyimpanan pada suhu 5 derajat C.

This study was aimed to determine the effect of additional of Curcuma xanthorrhiza extract on egg yolk citrate diluent towards the spermatozoa quality of thin-tailed ram. Fresh semen from thin-tailed ram aged 3 years was collected by an artificial vagina. Fresh semen was diluted using egg yolk citrate with the addition of Curcuma xanthorrhiza extract. The treatment consisted of egg yolk citrate as a control (P0), egg yolk citrate with the addition of Curcuma xanthorrhiza extract 1% (P1), 2% (P2), and 3% (P3). The diluted semen was stored in a refrigerator at 5 degree C. The variables observed in spermatozoa quality were motility (%), viability (%), and abnormality (%). The percentage of spermatozoa motility was determined subjectively and quantitatively. The viability and abnormality of spermatozoa were examined using eosin staining. Spermatozoa quality was evaluated after 3 hours and 3 days of storage. The obtained data were analyzed using One Way analysis of a completely randomized design, followed by Duncan Multiple Range Test. The results showed that the addition of Curcuma xanthorrhiza extract was significant differences (p<0.05) towards the motility, viability, and abnormality of spermatozoa after stored at 5 degree C. The quality of spermatozoa that observed after 3 hours stored showed that the addition of Curcuma xanthorrhiza extract 3% had the best result towards motility, viability, and abnormality of spermatozoa were 80,00+-2,00, 84,57+-1,10 and 7,82+-0,68% respectively. The addition of Curcuma xanthorrhiza extract 3% had the best result towards motility, viability, and abnormality of spermatozoa after 3 days stored were 61,67+-2,88, 77,54+-0,98 and 14,47+-0,90% respectively. The addition of 3% Curcuma xanthorrhiza extract gave the best semen quality after stored at 5 degree C.
