Optimasi Proses Homogenisasi Dalam Produksi Nanokapsul Karotenoid Arthrospira platensis Dengan Enkapsulan Gum Arab dan Whey Protein Concentrate

Arthrospira platensis merupakan salah satu golongan cyanobakteria yang mempunyai kandungan karotenoid sekitar 80% betakarotenoid dan dapat dikonversi menjadi vitamin A. Konsumsi karotenoid berhubungan erat dengan manfaat kesehatan diantaranya mengurangi resiko penyakit degeneratif. Sifat karotenoid yang mudah terisomerisasi dan teroksidasi menyebabkan tidak stabil dan mudah rusak di lingkungan, sehingga diperlukan proses nanoenkapsulasi. Proses enkapsulasi melibatkan homogenisasi yang berperan penting terhadap karakteristik fisik emulsi dan serbuk yang dihasilkan. Tujuan dari penelitian ini adalah untuk mengoptimasi proses homogenisasi dalam produksi kapsul ekstrak karotenoid A. platensis dengan enkapsulan gum arab dan whey protein concentrate menggunakan metode response surface methodology (RSM) dengan desain Box Behnken 15 run order. RSM mengevaluasi kombinasi lama waktu homogenisasi oleh high speed homogenizer, lama sonikasi dan amplitude yang digunakan sebagai parameter yang akan dioptimasi terhadap respon ukuran partikel serbuk, karotenoid total, karotenoid inti, efisiensi enkapsulasi, dan aktivitas antioksidan sebagai parameter utama dengan parameter pendukung berupa stabilitas emulsi, ukuran partikel emulsi, rendemen, karotenoid permukaan dan retensi karotenoid. Hasil penelitian menunjukkan kondisi optimal homogenisasi untuk mendapatkan ukuran partikel serbuk kecil (764,6 nm) dengan kandungan karotenoid total (99,71 mikrogram/mg db), karotenoid inti (71,97 mikrogram/mg db), efisiensi enkapsulasi (72,16%) dan aktivitas antioksidan tinggi (77,88%) adalah 90 detik high-speed homogenizer, 120 detik sonikasi dengan amplitude 82,5%, dengan nilai desirability 0,83.

Arthrospira platensis is a class of cyanobacteria that contains carotenoids around 80% betacarotenoids and can be converted to vitamin A. Consumption of carotenoids is closely related to health benefits including reducing the risk of degenerative diseases. Carotenoids is easily isomerized and oxidized, makes them unstable and easily damaged in the environment, so a nanoencapsulation process is needed. The encapsulation process involves homogenization which plays an important role in the physical characteristics of the emulsion and the resulting powder. The purpose of this study was to optimize the homogenization process in the production of A. platensis carotenoid extract capsules with Arabic gum and whey protein concentrate encapsulations using the response surface methodology (RSM) with Box Behnken 15 run order design. RSM evaluated the combination of homogenization time by a high-speed homogenizer, sonication time, and amplitude used as parameters to be optimized for the response of powder particle size, total carotenoids, core carotenoids, encapsulation efficiency, and antioxidant activity as the main parameters with supporting parameters in the form of emulsion stability, emulsion particle size, yield, surface carotenoids, and carotenoid retention. The results obtained in this study indicate that the optimal homogenization conditions to obtain small powder particle sizes (764,6 nm) with high total and core carotenoid content (99,71 microgram/mg db and 71,97 microgram/mg db, respectively), high encapsulation efficiency (72,16%) and high antioxidant activity (77,88%) are 90 seconds of high-speed homogenizer, 120 seconds of sonication with an amplitude of 82.5%, with a value of desirability 0,83.
