SINTESIS DAN KARAKTERISASI SIFAT KIMIAWI DAN FISIS COPPER NANOPARTICLES (CuNPs) DARI PREKURSOR Cu(NO3)2 SEBAGAI ZAT ANTIBAKTERI

Resistensi bakteri patogen terhadap antibiotik telah menjadi permasalahan dalam bidang kesehatan, Staphylococcus aureus merupakan salah satu bakteri patogen yang resisten methicillin. Bakteri dapat menyebar di udara atau melalui kontak fisik dengan penderita, sehingga masalah penyebaran strain resisten yang menyebabkan penyakit tidak dapat diselesaikan dengan mudah. Berdasarkan hal tersebut, diperlukan metode lain untuk mencegah penyebaran strain resisten dari rumah sakit agar tidak menginfeksi masyarakat. Copper nanoparticles (CuNPs) dapat digunakan sebagai desinfektan untuk pelapis peralatan non-sterile atau public service items. CuNPs dapat merusak membran sel dan membunuh bakteri patogen yang telah resisten terhadap antibiotik. Saat ini CuNPs komersial sering digunakan sebagai zat antibakteri, akan tetapi memiliki harga yang relatif mahal. Penelitian ini bertujuan untuk membandingkan kemampuan antibakteri dari CuNPs komersial dan CuNPs hasil sintesis menggunakan metode reduksi kimia dengan prekursor Cu(NO3)2. Metode reduksi kimia digunakan karena merupakan metode yang mudah dan efisien. Karakterisasi kimiawi dan fisis CuNPs dilakukan dengan menggunakan Scanning Electron Microscope (SEM), Particle Size Analyzer (PSA) dan Fourier Transform Infrared (FT-IR) Spectroscopy. Pengujian antibakteri dilakukan menggunakan teknik disc diffusion test, Alkaline Phosphatase Assay, dan membrane integrity staining. Hasil pengujian disc difufusion test menunjukkan CuNPs hasil sintesis (A1B1) lebih efektif menghambat pertumbuhan Escherichia coli FNCC 0091 sebesar 13,33 ± 0,58 mm dan Salmonella typhimurium FNCC 0134 sebesar 12,33 ± 0,58 mm, sedangkan CuNPs komersial (A1B2) lebih efektif menghambat pertumbuhan Staphylococcus aureus FNCC 0047 dengan diameter sebesar 23,7 ± 0,58 mm.

Resistance of pathogenic bacteria to antibiotics has become a problem in the health sector, Staphylococcus aureus is one of the methicillin-resistant pathogenic bacteria. Bacteria can be spread in the air or through physical contact with sufferers, so the problem of spreading resistant strains that cause disease cannot be solved easily. Based on this, other methods are needed to prevent the spread of resistant strains from hospitals so as not to infect the community. Copper nanoparticles (CuNPs) can be used as disinfectants for coating non-sterile equipment or public service items. CuNPs can damage cell membranes and kill pathogenic bacteria that are resistant to antibiotics. Currently, commercial CuNPs is often used as an antibacterial agent, but it has a relatively expensive price. This study aims to compare the antibacterial ability of commercial CuNPs and synthetic CuNPs using chemical reduction method with Cu(NO3)2 as a precursor. The chemical reduction method is used because it is an easy and efficient method. Chemical and physical characterization of CuNPs was carried out using Scanning Electron Microscope (SEM), Particle Size Analyzer (PSA) and Fourier Transform Infrared (FT-IR) Spectroscopy. Antibacterial testing was carried out using the disc diffusion test technique, Alkaline Phosphatase Assay, and membrane integrity staining. The results of the disc diffusion test showed that the synthesized CuNPs (A1B1) was more effective in inhibiting the growth of Escherichia coli FNCC 0091 by 13.33 ± 0.58 mm and Salmonella typhimurium FNCC 0134 by 12.33 ± 0.58 mm, while commercial CuNPs (A1B2) more effectively inhibited the growth of Staphylococcus aureus FNCC 0047 with a diameter of 23.7 ± 0.58 mm.
