IDENTIFIKASI FRAKSI PEPTIDA ANTIMIKROBA DARI HASIL HIDROLISIS TRIPSIN TERHADAP EKSTRAK TFA PROTEIN BIJI JARAK KEPYAR TERFRAKSINASI KOLOM PENUKAR KATION

Telah dilakukan penelitian dengan judul identifikasi fraksi peptida antimikroba dari hasil hidrolisis tripsin terhadap ekstrak TFA protein biji jarak kepyar terfraksinasi kolom penukar kation. Tujuan dari penelitian ini adalah untuk mengidentifikasi peptida yang bersifat antibakteri yang berasal dari hasil fraksinasi dan hidrolisat protein biji jarak kepyar dengan hidrolisis enzim tripsin Penelitian diawali dengan proses ekstraksi protein biji jarak kepyar dengan menggunakan asam kuat berupa TFA. Ekstrak kemudian dihidrolisis enzimatis dengan menggunakan enzim tripsin dan hidrolisat kemudian di fraksinasi dengan menggunakan kromatogrfi kolom penukar kation dielusi dengan variasi pH. Uji aktivitas antibakteri dilakukan dengan mengujikan hasil fraksinasi terhadap bakteri S.aureus dan E.coli menggunakan metode difusi cakram dan mikrodelusi. Fraksi peptida dengan aktivitas antibakteri diidentifikasi rantai peptidanya dengan menggunakan High Resolution Mass Spectrometry (HRMS). Hasil dari penelitian ini berupa ekstrak protein dengan rendemen sebesar 68,36% sedangkan hidrolisis enzimatis tripsin berhasil menghidrolisis 33,51% protein. Pengujian fraksi peptida dengan difusi cakram terhadap bakteri S.aureus dan E.coli menghasilkan zona hambat pada fraksi pH 3 dan pH 4. Nilai IC50 fraksi pH 3 dan 4 terhadap bakteri S.aureus berturut-turut adalah 4,25 ppm dan 17,96 ppm sedangkan nilai IC50 dari fraksi pH 3 dan 4 terhadap bakteri E.coli menghasilkan 22,50 ppm dan 25,74 ppm. Hasil identifikasi fraksi pH 3 dan 4 menggunakan HRMS menghasilkan 5 peptida dengan uturan asam amino AGVGIKPR, GAVVAPRK , ALQAPGR, GTPDDMGGAGAGNPPDDK dan AAGASG.

Research with the title of identification of antimicrobial peptide fraction from trypsin hydrolysis of TFA extract of jatropha seed protein fractionated by cation exchange column has been conducted. The purpose of this study was to identify peptides with antibacterial activity from the fractionation and hydrolyzation of jatropha seed protein with trypsin enzyme hydrolysis. The study began with the extraction of jatropha seed protein using a strong acid of TFA. The extract was then enzymatically hydrolyzed using trypsin and the hydrolyzate then fractionated using cation exchange column chromatography eluted with gradient pH. Antibacterial activity test was carried out by testing the fractions against S. aureus and E. coli bacteria using disc diffusion and microdelution methods. Peptide fractions with antibacterial activity were identified by using High Resolution Mass Spectrometry (HRMS). The protein extraction was succeeded to yield up to 68.36% while the enzymatic hydrolysis of trypsin succeeded in hydrolyzing 33.51% of protein. Testing the peptide fraction by disc diffusion against S. aureus and E. coli bacteria resulted in inhibition zones at the pH 3 and pH 4 fractions. The IC50 values of the pH 3 and 4 fractions against S. aureus bacteria were 4.25 ppm and 17.96 ppm, respectively, while the IC50 values of the pH 3 and 4 fractions against E. coli bacteria produced 22.50 ppm and 25.74 ppm, respectively.. The results of the identification of pH 3 and 4 fractions using HRMS produced 5 peptides with amino acid sequences AGVGIKPR, GAVVAPRK , GTPDDMGGAGAGNPPDDK, ALQAPGR and AAGASG.
