Karakter Fenotipik dan Polimorfisme Gen TYRP1 pada Ayam Hibrida F4 Golden Kamper (Gallus gallus domesticus Linnaeus, 1758)

Ayam Golden Kamper merupakan ayam hasil persilangan antara ayam jantan Pelung dengan betina Layer. Ayam ini memiliki warna dan corak bulu yang masih bervariasi. Hal tersebut membuat tujuan keseragaman warna bulu masih belum tercapai. Warna bulu yang diinginkan adalah blirik coklat-keemasan yang menjadi ciri khas dari jenis ayam ini. Salah satu warna bulu yang tidak diinginkan yang sering muncul di keturunan ayam Golden Kamper adalah warna bulu coklat. Pada ayam, adanya polimorfisme mutasi missense gen TYRP1 menyebabkan mutasi sex-linkage berupa warna bulu cokelat. Penelitian ini bertujuan untuk mengetahui karakter fenotipik khususnya warna bulu, mempelajari pengaruh polimorfisme gen TYRP1 pada F4 Golden Kamper, dan mengamati pertumbuhan ayam hingga umur 49 hari. Metode yang dilakukan adalah selective breeding sesama F3 Golden Kamper, koleksi telur F3 Golden Kamper, pemeliharaan Day Old Chick (DOC) F4 Golden Kamper, koleksi data fenotip, koleksi darah, isolasi DNA, amplifikasi DNA, sekuensing, elektroforesis, dan analisis data. Hasil penelitian menunjukkan F4 Golden Kamper memiliki karakter fenotipik berupa bentuk jengger single (100%); warna ceker/shank putih (31,25%), kuning (62,5%), dan abu-abu kehitaman (6,25%); serta warna bulu Blirik Coklat-Keemasan (12,5%), Blirik Putih-Keemasan (12,5%), Coklat (68,75%) dan Hitam-Coklat (6,25%). Bobot tubuh DOC F4 Golden Kamper pada umur 7 minggu mencapai 597,3 gram. Hasil morfometri menunjukkan bahwa F4 Golden Kamper memiliki postur dan proporsi tubuh menyerupai ayam Pelung, namun dengan bobot yang lebih tinggi. Sebanyak 14 polimorfisme substitusi ditemukan pada gen TYRP1 ayam F4 Golden Kamper. Berdasarkan Uji Korelasi Fisher's Exact Test, seluruh titik polimorfisme tersebut tidak memiliki korelasi terhadap kemunculan fenotip warna bulu coklat pada F4 Golden Kamper (p=1,00 ; p>0,05). Maka dari itu, gen TYRP1 belum dapat dijadikan sebagai penanda genetik fenotip warna bulu coklat, warna yang tidak diinginkan di keturunan ayam Golden Kamper.

Golden Kamper chicken is the result of a crossbreed between Pelung male and Layer female chicken. This chicken has a color and pattern of plumage that still vary. This makes the goal of uniformity plumage color not achieved. The desired plumage color is golden-brown blirik which is the hallmark of this chicken breed. One of the unwanted plumage colors that often appears in Golden Kamper breeds is brown. In chickens, the missense mutation polymorphism of the TYRP1 gene causes a sex-linkage mutation in the form of brown plumage. This study aimed to determine the phenotypic characters, especially plumage color, then to study the effect of the TYRP1 gene polymorphism on F4 Golden Kamper, and to observe the growth of chickens till 49 days of age. The methods used are, selective breeding among F3 Golden Kamper, collection of F3 Golden Kamper's eggs, treatment of Day Old Chick (DOC) F4 Golden Kamper, phenotypic data collection, blood collection, DNA isolation, DNA amplification, sequencing, electrophoresis, and data analysis. The results showed that F4 Golden Kamper had a phenotypic character in the form of a single comb (100%); the color of the shank are white (31.25%), yellow (62.5%), and blackish gray (6.25%); and color of plumage are Blirik Brown-Golden (12.5%), Blirik White-Golden (12.5%), Brown (68.75%) and Black-Brown (6.25%). The body weight of DOC F4 Golden Kamper at the age of 7 weeks reached 597.3 grams. The morphometric results showed that F4 Golden Kamper had the same posture and body proportions as Pelung chickens, but with a higher weight. A total of 14 substitution polymorphisms were found in the TYRP1 gene of chicken F4 Golden Kamper. Based on the Fisher's Exact Correlation Test, all Single Nuclotide Polymorphisms (SNPs) had no correlation with the appearance of the brown plumage color phenotype in F4 Golden Kamper (p=1.00 ; p>0.05). Therefore, the TYRP1 gene cannot be used as a genetic marker for the brown plumage color phenotype, an undesirable color in Golden Kamper breeds.
