UJI SENSITIVITAS BERBAGAI JENIS ANTIBIOTIK TERHADAP Staphylococcus aureus PADA SUSU SAPI MASTITIS

Salah satu penyebab rendahnya produksi susu pada sapi perah adalah adanya penyakit. Penyakit yang paling sering menyerang sapi perah adalah mastitis. Staphylococcus aureus merupakan salah satu agen utama penyebab mastitis. Penggunaan antibiotik sangat penting dalam pengobatan mastitis. Tingkat resistensi bakteri terhadap antibiotik yang semakin meningkat menyebabkan perlunya mengetahui antibiotik yang efektif untuk pengobatan penyakit. Penelitian ini bertujuan untuk mengetahui sensitivitas berbagai jenis antibiotik terhadap bakteri penyebab mastitis pada sapi. Sampel berupa susu sapi mastitis yang didapatkan dari hasil uji mastitis menggunakan reagen California Mastitis Test (CMT), reagen White Side Test (WST), reagen IPB-1, dan deterjen 5%. Susu kemudian dilakukan identifikasi Staphylococcus aureus menggunakan media Mannitol Salt Agar (MSA) dengan biakan murni Staphylococcus aureus sebagai kontrol positif. Staphylococcus aureus kemudian diuji sensitivitasnya terhadap antibiotik amoksisilin, ampisilin, kloramfenikol, eritromisin, gentamisin, penisilin, streptomisin dan oksitetrasiklin menggunakan metode Kirby-Bauer. Hasil uji sensitivitas dianalisis menggunakan analisa deskriptif. Uji mastitis dengan reagen CMT, WST, IPB-1, dan deterjen 5% menunjukkan hasil positif pada susu yang berasal dari ambing depan kanan. Identifikasi Staphylococcus aureus menunjukkan pertumbuhan koloni berwarna kuning pada media Mannitol Salt Agar (MSA) yang diinokulasi susu dari ambing depan kanan dan belakang kanan. Hasil uji sensitivitas antibiotik menunjukkan bahwa antibiotik kloramfenikol, eritromisin, dan gentamisin sensitif terhadap bakteri, namun antibiotik amoksisilin, ampisilin, penisilin, streptomisin, dan oksitetrasiklin resisten terhadap bakteri.

One of the causes of low milk production in dairy cows is the disease. The disease that most commonly attacks dairy cows is mastitis. Staphylococcus aureus is one of the main causative agents of mastitis. The use of antibiotics is very important for mastitis treatment. The increasing level of bacterial resistance to antibiotics drives the need to know which antibiotics are effective in treating the disease. This study aims to determine the sensitivity of various types of antibiotics against bacteria causing mastitis in cows. The sample was mastitis cow's milk obtained from mastitis test results using California Mastitis Test (CMT) reagent, White Side Test (WST) reagent, IPB-1 reagent, and 5%-detergent reagent. Staphylococcus aureus in milk was then identified using Mannitol Salt Agar (MSA) medium with a pure culture of Staphylococcus aureus as a positive control. Staphylococcus aureus were then tested for their susceptibility to amoxicillin, ampicillin, chloramphenicol, erythromycin, gentamicin, penicillin, streptomycin, and oxytetracycline using the Kirby-Bauer method. The results of the susceptibility test were analyzed using descriptive analysis. The mastitis test showed a positive result in the milk from the right fore udder. Identification of Staphylococcus aureus showed the growth of yellow colonies on Mannitol Salt Agar (MSA) media inoculated with milk from the right fore and rear udder. The results of the antibiotic susceptibility test showed that the antibiotics chloramphenicol, erythromycin and gentamicin were sensitive, but the antibiotics amoxicillin, ampicillin, penicillin, streptomycin and oxytetracycline were resistant to the bacteria.
