Pengaruh Campuran Glukosa dan Etanol Terhadap Pertumbuhan Dan Kandungan Alfa-Tocopherol Pada Kultur Fotoheterotrofik Euglena sp. Strain 2 Skala Laboratorium

Euglena merupakan mikroalga potensial karena diketahui menghasilkan Vitamin C dan E, protein, lipid, paramylon, serta memiliki biomassa yang tinggi. Walaupun mikroalga ini mudah ditemukan di perairan tawar, penelitian mengenai optimasi kultivasi Euglena di Indonesia masih jarang ditemukan. Tocopherol adalah antioksidan yang dapat mencegah berbagai macam penyakit yang disebabkan oleh kerusakan oksidatif. Isomer dari tocopherol yang memiliki aktivitas biologi paling tinggi adalah alfa-tocopherol. Kandungan alfa-tocopherol dalam sel Euglena sp. diketahui memiliki kadar paling tinggi dibandingkan dengan mikroorganisme dan mikeoalga lainnya. Hingga kini, peneliti masih mencari solusi untuk mendapatkan kandungan alfa-tocopherol yang tinggi sekaligus biomassa sel Euglena sp. yang tinggi. Kultur fotoautotrofik diketahui dapat meningkatkan kandungan tocopherol dalam Euglena, sedangkan kultur heterotrofik dapat meningkatkan biomassa. Maka dalam penelitian ini dilakukan kultur fotoheterotrofik dengan campuran glukosa dan etanol untuk meningkatkan kandungan alfa-tocopherol dan biomassa sekaligus dalam kultur Euglena sp. Pada penelitian ini, dilakukan penambahan variasi campuran glukosa dan etanol dengan konsentrasi berturut-turut 3:2; 2,5:2,5; 2:2; dan 0:0 (kontrol) g/L untuk mengetahui pengaruhnya terhadap pertumbuhan, biomassa dan kandungan alfa-tocopherol pada kultur Euglena sp. strain Indonesia. Hasil penelitian ini menunjukkan bahwa specific growth rate dan biomassa tertinggi didapatkan pada perlakuan 3:2 yaitu berturut-turut sebesar 0,992±0,049 (OD680/OD680/H) dan 8,48±0,28 (g/L), dan kandungan alfa-tocopherol tertinggi didapatkan pada perlakuan 2,5:2,5 yaitu sebesar 7,09±0,096 mg/L.

Euglena is a potential microalgae because it is known to contain vitamins C and E, protein, lipid, and paramylon and additionally has larger biomass content. Although these microalgae are easily found in freshwater, research on the optimization of Euglena cultivation in Indonesia is still rare. Tocopherols are antioxidants that can prevent a wide variety of diseases caused by oxidative damage. The isomer of tocopherol which has the highest biological activity is alpha-tocopherol. The content of alpha-tocopherol in Euglena's cells is known to have the highest levels compared to other microorganisms such as yeasts, molds and macroalgae. Until now, researchers are still working for a solution to obtain high alpha-tocopherol content as well as high Euglena cell biomass. Photoautotrophic culture is known to increase the tocopherol content in Euglena, while heterotrophic culture could increase biomass. Therefore, in this study photoheterotrophic culture with a mixture of glucose and ethanol was carried out to increase the alpha-tocopherol and biomass content in the culture of Euglena sp. Indonesian strain. In this study, the addition of variations in the mixture of glucose and ethanol with concentrations of 3:2; 2.5: 2.5; 2: 2; and 0: 0 (control) g/L, respectively, to determine the effect on growth, biomass and alpha-tocopherol content in the culture of Euglena sp. The results of this study indicate that the highest specific growth rate and biomass was obtained in the 3:2 treatment, namely 0.992±0.049 (OD680/OD680/day) and 8.48±0.28 (g/L), and the highest alpha-tocopherol content was obtained in 2.5: 2.5 treatment, namely the amount of 7.09±0.096 mg/L.
