Potensi Prebiotik dan Identifikasi Molekuler Rumput Laut Merah (Rhodophyta) dari Pantai Selatan Kabupaten Gunungkidul

Rumput laut merah (Rhodophyta) Indonesia sangatlah melimpah dan beragam. Banyak spesies rumput laut merah penghasil polisakarida dari pantai di kabupaten Gunungkidul yang belum dieksplorasi dan diidentifikasi. Salah satu potensi rumput laut merah adalah sebagai penghasil polisakarida dan prebiotik. Penelitian ini bertujuan untuk mendapatkan polisakarida dari rumput laut merah dari Gunungkidul yang dapat dimanfaatkan sebagai prebiotik untuk akuakultur. Selanjutnya melakukan identifikasi molekuler untuk species rumput laut merah yang berpotensi. Polisakarida rumput laut diperoleh dengan metode ekstraksi pengendapan dengan etanol dingin. Identifikasi jenis polisakarida dilakukan dengan cara menganalisis gugus fungsional karbohidrat dan sidik jari senyawa berdasarkan spektra FT-IR (Fourir-transform infrared spectroscopy). Pengujian pengaruh polisakarida terhadap pertumbuhan probiotik dilakukan dengan cara pengukuran pertumbuhan probiotik pada medium minimal. Uji ketahanan asam polisakarida dilakukan merendam dalam larutan asam pH 5 selama 4-6 jam kemudian dilakukan analisis Kromatografi lapis tipis. Identifikasi molekuler rumput laut potensial sebagai prebiotik dilakukan menggunakan gen target COI berdasarkan metode Polymerase Chain Reaction (PCR). Hasil penelitian menunjukkan jenis polisakarida sampel A (DR-01), B (DR-02), dan C (NR-05) adalah kelompok galaktan tersulfatasi. Ketiga jenis polisakarida tersebut dapat bertahan pada keasaman pH 5 selama 4-6 jam. Galaktan sulfat paling potensial sebagai prebiotik dihasilkan oleh rumput laut merah B (DR-01). Identifikasi molekuler rumput laut potensial B (DR-01) memiliki tingkat kemiripan dengan rumput laut Gracilaria edulis sebesar (99,68%).

Red seaweed (Rhodophyta) Indonesia is very abundant and diverse. Many species of red seaweed that produce polysaccharides from the coast in Gunungkidul have not been explored and identified. Red seaweeds are potential as producer of polysaccharides and prebiotics. This study aims to obtain polysaccharides from red seaweed from Gunungkidul which can be used as prebiotics for aquaculture. Furthermore, molecular identification of potential red seaweed species is carried out. Seaweed polysaccharides were obtained by precipitation in cold absolute ethanol. Identification of the type of polysaccharide was carried out by analyzing the functional groups of carbohydrates and the fingerprints of the compound based on the FT-IR (Fourir-transform infrared spectroscopy) spectra. The examination of the effect of polysaccharides on the growth of probiotics was carried out by measuring the growth of probiotics in minimal medium in the presence of each polysaccharide. The acid resistance test of polysaccharides was carried out by immersing in an acid solution of pH 5 for 4-6 hours, then thin layer chromatography analysis was performed. Molecular identification of potential seaweeds as prebiotics was carried out using COI target genes based on the Polymerase Chain Reaction (PCR) method. The results showed that the types of polysaccharide samples A (DR-01), B (DR-02), and C (NR-05) were sulfated galactan groups. The three types of polysaccharides were stable at pH 5 for 4-6 hours. The most potential sulfate galactan as a prebiotic is produced by red seaweed B (DR-01). Molecular identification of potential seaweed B (DR-01) has a similarity with Gracilaria edulis (99.68%).
