PENGARUH PEMBERIAN DEKOKTA TEH HIJAU (Camellia sinensis) TERHADAP TITER HEMAGLUTINASI DAN PERUBAHAN MAKROSKOPIK EMBRIO TELUR AYAM BERTUNAS YANG DIINOKULASI VIRUS VAKSIN NEWCASTLE DISEASE

Newcastle Disease (ND) merupakan penyakit endemik yang menyerang unggas dengan gejala klinis depresi, paralisis, tortikolis, dan gejala pernafasan. Obat spesifik dapat mengatasi penyakit yang disebabkan virus ND hingga saat ini belum ditemukan. Vaksinasi dilakukan sebagai salah satu pencegahan terhadap virus ND, namun penggunaan vaksin aktif dapat menimbulkan reaksi pasca vaksinasi dan pada vaksin inaktif rangsangan imunitas muncul lebih lama, sehingga diperlukan alternatif untuk meminimalisir dampak penyakit, yaitu dengan penggunaan bahan herbal. Teh hijau (Camellia sinensis) mengandung senyawa catechin yang terdiri dari epigallocatechin, epicatechin-gallate, epicatechin. Senyawa-senyawa tersebut memiliki aktivitas antiviral dengan menghambat enzim RNA-dependen RNA polimerase pada replikasi virus dan disrupsi membran sel virus. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian dekokta teh hijau pada telur ayam bertunas (TAB) yang diinfeksi virus vaksin ND terhadap titer hemaglutinasi dan perubahan makroskopik embrio. Penelitian ini menggunakan 15 TAB umur 9 hari yang dibagi menjadi tiga kelompok yaitu kelompok kontrol positif (A), kelompok pemberian dekokta teh hijau konsentrasi 10% (B), dan 20% (C) yang diberi perlakuan sesuai masing-masing kelompok, diamati perubahan makroskopiknya 5 hari pasca perlakuan (umur 14 hari), dilanjutkan dengan pemeriksaan titer HA antigen dari cairan allantois berdasarkan uji hemaglutinasi. Hasil penelitian pada kelompok A rataan titer antigen virus sebesar 2 5,4 unit HA dengan embrio menunjukkan hemoragi diseluruh tubuh, kerdil, dan tidak ada pertumbuhan bulu, pada kelompok B rataan titer antigen virus 2 2,6 unit HA dengan embrio terlihat kongesti ringan, kerdil dan tidak ada pertumbuhan bulu, pada kelompok C rataan titer antigen virus 2 0,8 unit HA dengan embrio teramati normal (tidak terjadi hemoragi dan ada pertumbuhan bulu). Berdasar analisa statistik dengan uji one way ANOVA dilanjut dengan uji berjarak Duncan ada perbedaan signifikan antara kelompok C dengan kelompok A. Kesimpulan dari penelitian ini pemberian dekokta teh hijau konsentrasi 20% memberikan pengaruh yang signifikan terhadap penurunan titer antigen virus dan perubahan makroskopik embrio TAB yang diinokulasi virus ND.

Newcastle Disease (ND) is an infectious endemic disease of birds with clinical signs found are depression, paralyze, torticollis, and respiratory symptoms. The spesific treatment for disease caused by the ND virus still not available. Vaccination is also carried out as a preventive measure against the ND virus, however the use of live vaccines can result in post vaccination reaction and for the killed vaccines, the stimulation of immunity to appear longer, so an alternative is needed to minimize the impact of the disease, one of them is using alternative from herbal ingredients. Green tea (Camellia sinensis) contains catechin compounds consist of epigallocatechin (EGC), epicatechin-gallate (ECG), epicatechin (EC). These compounds have an antiviral activity that plays a role on inhibiting RNA-dependent RNA polymerase enzymes involved in viral replication and disruption of viral cell membranes. This study aims to determine the effect of green tea decoct to embryonated chicken eggs (TAB) infected with ND vaccine virus on haemagglutination titre and macroscopic changes. This study used 15 TAB 9-day-old which were divided into three groups, the group as positive control (A), the group treated decoct of green tea 10% (B) and 20% (C) and treated according to each group, observed the macroscopic changes 5 days after treatment (14-day-old), followed by examination of the HA antigen titre of allantoic fluid based on the hemagglutination test. The results of the study in groups A mean viral antigen titre was 2 5,4 HA units with the embryo showed hemorrhage throughout the body accompanied by stunted embryos, and no hair growth, in group B viral mean antigen titre was 2 2,6 HA units with the embryo showed mild congestion on the skin, stunted embryos, and no hair growth, while in groups C mean viral antigen titre was 2 0,8 HA units with the embryo were observed normal growth (no hemorrhage and there is hair growth). Based on statistical analysis with one way ANOVA followed Duncan test there was a significant difference between group C and group A. The conclusion of this study, embryonated egg treated with green tea decoct with 20% concentration had a significant effect on decreasing viral antigen titre and macroscopic changes in embryonated egg inoculated with Newcastle Disease virus.
