Kajian Toksoplasmosis secara Molekuler pada Ular Kobra Jawa (Naja naja sputtatrix)

Toksoplasmosis merupakan penyakit parasiter zoonosis yang penularannya melalui rute oral dan dapat menginfeksi berbagai jenis hewan termasuk ular. Ular kobra jawa banyak dikonsumsi manusia dalam kondisi mentah (darah, sumsum tulang) dan setengah matang (daging). Penelitian ini bertujuan untuk mengkaji kejadian toksoplasmosis pada ular kobra jawa (Naja naja sputtatrix) berdasar uji molekuler. Sepuluh (10) sampel otak kobra jawa digunakan dalam penelitian ini. Sampel otak diperoleh dari tempat pemotongan ular kobra di jalan Imogiri Bantul Yogyakarta dan ular-ular tersebut tangkapan liar dari wilayah Gunung Kidul. Metode pemeriksaan menggunakan Polymerase Chain Reaction (PCR) konvensional dengan primer gen B1 dan ukuran amplikon 555 bp. Hasil uji PCR diperoleh data semua sampel negatif toksoplasmosis. Hasil negatif diduga berhubungan dengan jumlah sampel, asal-usul sampel dan metode pemeriksaan terutama jenis primer yang digunakan. Dapat disimpulkan bahwa ular kobra jawa penelitian negatif toksoplasmosis namun secara ilmiah, ular dapat tertular toksoplasmosis. Perlu penelitian lebih lanjut dengan jumlah sampel yang lebih banyak dan variasi lokasi sampel serta penggunaan primer tipe GRA6 sebagai pembanding hasil.

Toxoplasmosis is a zoonotic parasitic disease which is transmitted by oral route and can infect various types of animals, including snakes. The Javan cobra is mostly consumed by humans in raw conditions (blood, bone marrow) and half cooked (meat). This study aims to examine the incidence of toxoplasmosis in the Javan cobra (Naja naja sputtatrix) based on molecular tests. Ten (10) Javan cobra brain samples were used in this study. Brain samples were obtained from cobra slaughterhouses on Jalan Imogiri, Bantul, Yogyakarta and the snakes were caught wild from the Gunung Kidul area. The examination method used conventional Polymerase Chain Reaction (PCR) with B1 gene primers and 555 bp amplicon size. The PCR test results showed that all samples were negative for toxoplasmosis. The negative results are thought to be related to the number of samples, the origin of the samples and the method of examination, especially the type of primer used. It can be concluded that the Javan cobra is negative for toxoplasmosis, but scientifically, snakes can contract toxoplasmosis. Further research is needed with a larger number of samples and variations in sample locations and the use of GRA6-type primers as a comparison of results.
