PENGARUH WAKTU SIMPAN SIMPLISIA KUNCUP BUNGA CENGKIH TERHADAP PROFIL GC-MS MINYAK ATSIRI DAN POTENSI SITOTOKSIK PADA SEL 4T1

Cengkih (Syzigium aromaticum L.) adalah salah satu tanaman asli Indonesia yang berasal dari daerah Maluku. Proses pascapanen dapat memengaruhi kandungan kimia di dalam cengkih, sehingga proses ini merupakan faktor kritis dalam menentukan profil fitokimia minyak atsiri yang akan dihasilkan. Komposisi minyak atsiri yang berbeda-beda mengakibatkan kemungkinan perbedaan efek yang dihasilkan. Penelitian ini bertujuan untuk mengetahui perbedaan profil fitokimia minyak cengkih yang memiliki perbedaan pada masa penyimpanan dan efeknya terhadap aktivitas sitotoksik pada sel kanker. Bahan kuncup bunga cengkih kering dengan 3 (tiga) waktu penyimpanan berbeda diperoleh dari petani di daerah Boyolali, didistilasi dengan teknik distilasi uap-air untuk mendapatkan minyak cengkih. Minyak tersebut kemudian dikarakterisasi dengan gas chromatography-mass spectrometry (GC-MS) untuk mendapatkan profil kandungan senyawa minyak atsirinya. Evaluasi aktivitas sitotoksik masing-masing minyak dilakukan terhadap sel kanker payudara 4T1 dengan MTT assay dengan luaran IC50. Distilasi minyak cengkih (MC) dari kuncup bunga cengkih kering dengan waktu penyimpanan berbeda yaitu 1 tahun, 4 tahun, dan 6 tahun menghasilkan jumlah rendemen yang berbeda-beda yaitu 2,53%, 5,61%, dan 9,95%. Semakin lama disimpan, minyak yang dihasilkan semakin banyak. Profil fitokimia GC-MS menunjukkan minyak dari cengkih dengan waktu penyimpanan berbeda memiliki komposisi senyawa yang berbeda-beda. Perbedaannya terdapat pada senyawa guaiacol, eugenol, 1-caryophyllene, dan beberapa senyawa lainnya. Minyak dari cengkih dengan waktu penyimpanan lebih singkat (1 tahun) memiliki aktivitas sitotoksik lebih kuat (100 ug/mL) dibanding dengan minyak dari cengkih dengan waktu penyimpanan 4 tahun (178 ug/mL) dan 6 tahun (175 ug/mL). Penelitian ini menunjukkan bahwa cengkih yang disimpan dengan waktu yang berbeda-beda menghasilkan minyak dengan profil fitokimia yang berbeda serta aktivitas yang berbeda pula.

Cloves (Syzigium aromaticum L.) is one of Indonesia's native plants originating from the Moluccas region. The post-harvest process can affect the chemical content in cloves, so this process is a critical factor in determining the quality of the essential oil that will be produced. The different essential oil compositions may result in different effects. This study aims to determine differences in the phytochemical profiles of clove oil which have different storage periods and their effects on cytotoxic activity in cancer cells. Dried clove flower buds with 3 (three) different ages were obtained from farmers in the Boyolali area, distilled by the water-steam distillation technique to obtain clove oil. The oil is then characterized by gas chromatography-mass spectrometry (GC-MS) to profile its essential oil compound content. The evaluation of each oil's cytotoxic activity was carried out on breast cancer 4T1 cells using the MTT assay method with an IC50 output. Distillation of clove oil from dried clove flower buds with different ages (1 year, 4 years, and 6 years) resulted in different yields by 2,53%, 5,61%, and 9,95% respectively, indicated that the longer the storage of the oil, the more oil would be produced. The GC-MS phytochemical profile shows that oil from cloves with different ages had different compound compositions, including guaiacol, eugenol, and 1-caryophyllene. The oil from cloves with shortest storage time (1 year) has strongest cytotoxicity activity (100 ug/mL) than oil from clover with 4 years (178 ug/mL) and 6 years (175 ug/mL) of storage time. The result above reveals that cloves stored for different times produce oil with different qualities and different cytotoxic activities.
