Potensi Minyak Akar Wangi (Vetiveria zizanioides L.) sebagai Agen Sitotoksik pada Sel Kanker Triple Negative Breast Cancer (TNBC) 4T1 melalui Penghambatan Siklus Sel

Penelusuran agen sitotoksik dari bahan alam dilakukan untuk mengembangkan strategi terapi yang efektif dan aman pada triple negative breast cancer (TNBC). Minyak akar wangi (Vetiveria zizanioides L.) (MAW) mengandung senyawa-senyawa seskuiterpen, seperti beta-caryophyllene (BCP) dan alfa-humulene, yang diketahui sitotoksik pada sel kanker dan menghambat siklus sel. Penelitian ini bertujuan untuk mengungkap potensi MAW sebagai agen sitotoksik melalui penghambatan siklus sel dan mempengaruhi level reactive oxygen species (ROS) pada sel kanker payudara 4T1 sebagai model TNBC. Minyak akar wangi (MAW) diperoleh melalui metode distilasi uap air. Karakterisasi kandungan senyawa MAW dilakukan menggunakan gas chromatography-mass spectrometry (GC-MS). Efek sitotoksik MAW pada sel 4T1 ditentukan dengan MTT assay. Efek MAW terhadap penghambatan siklus sel pada 4T1 diukur dengan uji siklus sel menggunakan flowcytometry. Selain itu, efek MAW terhadap level ROS intraseluler diukur dengan DCFDA staining assay. Hasil karakterisasi senyawa menunjukkan bahwa MAW mengandung komponen senyawa utama alfa-vetivone, beta-vetivone, beta-caryophyllene, dan alfa-humulene. Uji sitotoksik menunjukkan bahwa MAW bersifat sitotoksik kuat pada sel 4T1 dengan IC50 sebesar 58,9 �µg/mL. Uji siklus sel menunjukkan bahwa perlakuan MAW pada dosis sekitar IC50 mampu menghambat siklus sel pada fase G2/M. Sementara itu, perlakuan MAW tidak berpengaruh terhadap level ROS intraseluler. Dengan demikian, MAW berpotensi untuk dikembangkan sebagai agen sitotoksik pada model TNBC melalui penghambatan siklus sel.

The exploration of cytotoxic agent from natural ingredients has been carried out to develop effective and safe therapeutic strategies for triple negative breast cancer (TNBC). Vetiver root (Vetiveria zizanioides L.) oil contains sesquiterpene compounds, such as beta-caryophyllene (BCP) and alpha-humulene, which are cytotoxic and able to induce cell cell cycle arrest. This study aims to reveal the potency of vetiver root oil (VO) as a cytotoxic agent through cell cycle arrest and reactive oxygen species (ROS) level on 4T1 breast cancer cells. Vetiver root oil (VO) was obtained through the steam water distillation. Characterization of VO composition was performed using gas chromatography-mass spectrometry (GC-MS). VO cytotoxicity against 4T1 cells was determined based on the cell viability by MTT assay. The effect of VO on cell cycle inhibition in 4T1 was assessed by cell cycle assay using flowcytometry. Meanwhile, the VO effect on intracellular ROS level was measured by DCFDA staining assay The characterization showed that VO contained main components as follows: alpha-vetivone, beta-vetivone, beta-caryophyllene (BCP), and alpha-humulene. Cytotoxic tests showed that VO had a strong cytotocixiy toward 4T1 cells with an IC50 of 58,9 �¼g/mL. Furthermore, VO at around the IC50 arrested cell cycle on G2/M phase. Meanwhile, VO treatment had no effect in ROS level. Therefore, VO is potential to be developed as a cytotoxic agent for TNBC trough cell cycle arrest.
