SIMULTANEOUS DETERMINATION OF SEVEN FOOD ADDITIVES AND CAFFEINE IN POWDERED DRINKS USING HPLC DAD: METHOD DEVELOPMENT AND VALIDATION

Tujuan dari penelitian ini adalah untuk mengembangkan dan memvalidasi metode analisa Kromatografi Cair Kinerja Tinggi dengan Detektor Diode Array (HPLC-DAD) untuk penentuan secara simultan tujuh bahan tambahan pangan dan kafein dalam minuman serbuk berperisa berdasarkan pada Response Surface Methodology (RSM) menggunakan pendekatan Box Behnken Design (BBD). Kombinasi tujuh bahan tambahan pangan dan kafein (asesulfam kalium (ACE), asam benzoat (BEN), asam sorbat (SOR), natrium sakarin (SAC), tartrazin (TAR), kafein (CAF), sunset yellow FCF (SUN), dan aspartam (ASP)) dianalisa dengan menggunakan sistem KCKT Shimadzu (Kyoto, Jepang) yang dilengkapi dengan pompa biner (LC-20AD), diode array detector (DAD) (SPD M20A), dan autosampler (SIL-HTC, Shimadzu, Jepang) . Pemisahan yang berhasil dicapai dengan eluasi secara gradien pada kolom C18 fase terbalik Shim-Pac GIST Shimadzu (150 mm, 4,6 mm, 5 mikro meter), Fase gerak yang digunakan adalah buffer fosfat dalam air (pH 3, 5, dan 7) dan metanol dioptimalkan % B awal 0% hingga 10% dan % B akhir 60% hingga 100% diikuti oleh laju alir 1,0 mL /menit. Panjang gelombang deteksi berada pada 210 nm. Metode optimasi kromatografi diperoleh buffer fosfat fase gerak dalam air (pH 6,7)% B awal 8,5% dan % B akhir 90% untuk tujuh bahan tambahan pangan dan kafein. Untuk delapan analit ini, hasilnya menunjukkan bahwa uji kesesuaian sistem (% RSD test) kurang dari <2%, batas deteksi adalah 1,16 hingga 3,00 mg/Kg, dan batas kuantifikasi adalah 3,86 hingga 10,02 mg/Kg. Konsentrasi linearitas adalah 1-50 mg/L. Koefisien determinasi ditemukan lebih besar dari 0,9990. Simpangan baku relatif antar hari dan antar hari kurang dari 4,06%. Pemulihan adalah 95,30 hingga 101,74%. Metode ini menyediakan teknis yang diperlukan untuk kuantifikasi tujuh bahan tambahan pangan dan kafein yang dipilih untuk pengujian. The objective of this study was to develop and to validate a high-performance liquid chromatography diode array detector (HPLC-DAD) method for the simultaneous determination of seven food additives and caffeine in instant drink flavored powder based on Response Surface Methodology using Box Behnken Design (BBD) approach. The combination of seven food additives and caffeine (acesulfame potassium (ACE), benzoic acid (BEN), sorbic acid (SOR), sodium saccharin (SAC), tartrazine (TAR), caffeine (CAF), sunset yellow FCF (SUN), and aspartame (ASP)) was analyzed by using Shimadzu HPLC system (Kyoto, Japan) equipped with binary pump (LC-20AD), diode array detector (DAD) (SPD M20A), and autosampler (SIL-HTC, Shimadzu, Japan). The successful separation was achieved by gradient elution on a reverse-phase C18 column Shim-Pac GIST Shimadzu(150 mm, 4.6 mm, mikrometer), The mobile phase used was phosphate buffer in water (pH 3, 5, and 7) and methanol on was optimized %B initial 0%-10% and %B end 60%-100% followed by 1.0 mL/min flow rate. The wavelength of detection was at 210 nm. The chromatographic optimum method was the mobile phases phosphate buffer in water (pH 6.7) %B initial 8.5% and %B end 90% for seven food additives and caffeine. For these eight analytes, the results showed that the system suitability tests (%RSD test) were less than <2%, the limits of detection were 1.16 to 3.00 mg/Kg, and the limits of quantification were 3.86 to 10.02 mg/Kg. The linearity concentrations were 1-50 mg/L. The coefficients of determination were found to be greater than 0.999. The relative standard deviations of inter- and intra-day were less than 4.06%. The Recoveries were 95.30 to 101.74 %. This method provides a necessary tool for the quantification of the selected seven food additives and caffeine for their assay. The proposed method is simple, accurate, reproducible, and applied successfully to analyze them in powdered drinks.
