Potensi Sitotoksik Kombinasi Minyak Bekatul (Oryza Sativa L.) dengan Doksorubisin melalui Pemacuan Apoptosis pada Sel Kanker Payudara TNBC, 4T1

Timbulnya efek samping dan kemoresistensi akibat doksorubisin pada kanker mendorong pengembangan agen peningkat efektivitas terapi dari bahan herbal. Minyak bekatul diketahui mengandung senyawa-senyawa yang memiliki aktivitas antioksidan dan sitotoksik terhadap sel kanker. Penelitian ini bertujuan untuk menelusuri potensi minyak bekatul (BKT) sebagai agen ko-kemoterapi doksorubisin melalui pemacuan apoptosis pada model sel kanker 4T1 yang termasuk subtipe Triple Negative Breast Cancer (TNBC). Minyak bekatul diekstraksi dengan n-heksana dan dikarakterisasi melalui pemisahan dengan High Performance Liquid Cromatography (HPLC). Aktivitas sitotoksik tunggal BKT dan kombinasi doksorubisin ditetapkan berdasarkan IC50 dan Combination Index melalui MTT Assay. Sementara itu, efek penghambatan daur sel dan pemacuan apoptosis diukur dengan flow cytometry setelah dilakukan pengecatan dengan Propidium Iodide (PI) dan Annexin-V/PI. Hasil penelitian menunjukkan bahwa BKT tidak bersifat sitotoksik pada sel 4T1 dengan IC50 450 ug/mL. Lebih lanjut, kombinasi BKT dengan doksorubisin bersifat sinergis dan secara signifikan (p<0,05) mampu menurunkan viabilitas sel dibanding doksorubisin. Perlakuan BKT konsentrasi 100 dan 200 µg/mL kombinasi doksorubisin secara signifikan mampu meningkatkan akumulasi sel fase sub-G1 dan dikonfirmasi dengan adanya induksi apoptosis pada perlakuan yang sama. Penelitian ini menunjukkan bahwa BKT berpotensi untuk dikembangkan sebagai agen ko-kemoterapi doksorubisin pada kanker payudara TNBC.

The side effect of doxorubicin and chemoresistance in cancer therapy encourage the development of therapeutic enhancing agent from herbal materials. Rice bran is known to contain compounds with antioxidant and cytotoxic activities against cancer cells. This study aims to explore the potential cytotoxic activity of rice bran oil (RBO) in single and combination with doxorubicin through apoptosis induction on 4T1 breast cancer cell model, which one of Triple Negative Breast Cancer (TNBC) subtype. Rice bran oil was extracted with n-hexane and was characterized through High Performance Liquid Cromatography (HPLC). RBO cytotoxic activity and combination with doxorubicin were determined based on IC50 and CI by MTT assay. Meanwhile, cell cycle inhibition and apoptosis induction were measured by flow cytometry after Propidium Iodide (PI) and Annexin-V/PI staining. The result showed that RBO is not toxic to 4T1 cells with IC50 of 450 µg/mL. Its combination with doxorubicin work synergist and significantly (p<0,05) decrease cell viability compared with doxorubicin. Treatment of RBO 100 dan 200 µg/mL combination with doxorubicin significantly increased cell accumulation in sub-G1 phase and it was confirmed by apoptosis induction in the same treatment. Based on these data, RBO has potential to be developed as co-chemotherapeutic agent with doxorubicin in TNBC breast cancer.
