ANALISIS REKOMBINAN KLON GEN PENYANDI PENISILIN G-ASILASE DARI ESCHERICHIA COLI DALAM INANG ESCHERICHIA COLI DH5ALFA

Tingginya angka infeksi di Indonesia belum diimbangi dengan kemandirian produksi antibiotik domestik. Untuk mengurangi ketergantungan impor antibiotik diperlukan upaya produksi bahan baku antibiotik secara mandiri, terutama antibiotik yang berspektrum luas diantaranya yaitu amoksisilin. Jalur biosintesis amoksisilin melibatkan reaksi enzimatik dengan enzim PGA sebagai enzim kunci sehingga produksi enzim PGA perlu dilakukan. Penelitian ini merupakan salah satu rangkaian dalam penelitian produksi enzim PGA menggunakan pendekatan DNA rekombinan. Enzim PGA yang ingin diproduksi berasal dari inang alami E. coli yang telah mengalami optimasi kodon pada gen penyandinya guna mengoptimalkan ekspresi enzim dalam sel inang E. coli BL-21(DE3) dan telah diinsersikan dalam plasmid ekspresi pET22b. Untuk mendapatkan enzim PGA rekombinan tersebut, salah satu tahap yang perlu dilakukan dan menjadi fokus dalam penelitian ini adalah perbanyakan rekombinan gen pga E. coli (pgaEc) sintetik dalam inang E. coli DH5alfa serta analisis rekombinan klon gen pgaEc menggunakan PCR dan sekuensing. Analisis ini bertujuan untuk mengidentifikasi kebenaran orientasi insersi rekombinan klon gen pgaEc. PCR dilakukan menggunakan dua primer, yaitu T7 forward dan reverse spesific primer (berturut-turut, yaitu 5-TAATACGACTCACTATAGGG-3 dan 5-GTACCAACAAAGATCATCG-3). Hasil elektroforesis terhadap produk PCR menunjukkan bahwa semua sampel plasmid yang diisolasi dari tiga koloni E. coli DH5alfa transforman positif mengandung gen pgaEc dengan orientasi insersi yang benar. Sementara itu, sekuensing juga dilakukan menggunakan primer yang sama. Hasil sekuensing memperkuat kebenaran orientasi insersi rekombinan klon gen pgaEc.

The high rate of infection in Indonesia has not been matched with the independence of domestic antibiotic production. To reduce the dependence of antibiotic import, independent efforts are needed to produce antibiotic raw materials, especially broad-spectrum antibiotics including amoxicillin. Amoxicillin biosynthesis pathway involves enzymatic reactions with PGA enzyme as a key enzyme so PGA enzyme production needs to be carried out. This research is part of studies on PGA enzymes production using recombinant DNA approach. PGA enzymes to be produced are derived from natural E. coli hosts that have undergone codon optimization in their encoding genes to optimize enzyme expression in E. coli BL-21(DE3) host cells and have been inserted in pET22b expression plasmids. To get the recombinant PGA enzyme, one of steps that needs to be done and become the focus of this research is the propagation of recombinant synthetic pga E. coli gene (pgaEc) in E. coli DH5alfa host and analysis of the recombinant clone using PCR and sequencing. The aim of this analysis is to identify the orientation of the pgaEc gene recombinant clone insertion. PCR was performed using two primers, namely T7 forward primer and reverse specific primer (respectively, 5-TAATACGACTCACTATAGGG-3 and 5-GTACCAACAAAGATCATCG-3). Electrophoresis result of PCR products showed that all plasmid samples isolated from three positive E. coli DH5α transformant colonies contain the pgaEc gene with the correct orientation of insertion. Meanwhile, sequencing was also carried out using the same primer. The sequencing results confirm the correct orientation of the pgaEc gene recombinant clone insertion.
