PENGARUH EKSTRAK DAUN KELOR (Moringa oleifera) TERHADAP PERLEKATAN BAKTERI Streptococcus mutans ATCC 25175 in vitro

Streptococcus mutans merupakan salah satu mikroorganisme yang memicu terjadinya karies gigi. Bakteri ini memiliki kemampuan melekat pada permukaan gigi. Ekstrak daun kelor (Moringa oleifera) mengandung bromelain, tanin, dan flavonoid. Kandungan tersebut kemungkinan dapat mempengaruhi kemampuan bakteri melekat pada suatu permukaan. Tujuan penelitian ini untuk mengetahui pengaruh ekstrak daun kelor terhadap kemampuan perlekatan bakteri Streptococcus mutans ATCC 25175. Metode penelitian menggunakan microtiter plate static biofilm assay. Bahan penelitian berupa ekstrak daun kelor konsentrasi 2,08%; 4,17%; dan 8,34% sebagai perlakuan, klorheksidin glukonat 0,2% sebagai kontrol positif dan akuades sebagai kontrol negatif. Sumuran diisi 50 µL ekstrak, klorheksidin, maupun akuades, kemudian ditambahkan BHI 40 µL, lalu diinokulasikan 10 µL suspensi bakteri 1,5 x 108 CFU/ml. Setiap kelompok dilakukan replikasi 6 kali. Microplate diinkubasi pada suhu 37 derajat celcius selama 24 jam menggunakan microplate reader pada panjang gelombang 540 nm. Analisis statistik menggunakan ANOVA dan Tukey pada p<0,05. One way ANOVA menunjukkan perbedaan signifikan pada persentase penghambatan antar kelompok, hal tersebut menunjukkan bahwa ekstrak daun kelor mempengaruhi kemampuan perlekatan S. mutans ATCC 25175. Post hoc Tukey HSD menunjukkan semua konsentrasi ekstrak daun kelor berbeda signifikan dibandingkan klorheksidin glukonat 0,2% dalam menghambat perlekatan S. mutans ATCC 25175. Kesimpulan penelitian ini adalah ekstrak daun kelor dapat menghambat perlekatan bakteri S. mutans ATCC 25175. Konsentrasi yang paling efektif dalam menghambat perlekatan S. mutans ATCC 25175 pada penelitian ini adalah 8,34%, walaupun secara bermakna masih di bawah potensi klorheksidin 0,2%.

Streptococcus mutans is a bacteria that triggers dental caries. This bacteria is able to adhere to the surfaces of the teeth. Moringa leaf extract contains bromelain, tannins, and flavonoids. These contents likely affect bacteria ability to adhere to a surface. The purpose of this study to determine the effect of Moringa leaf extract on the adhesion ability of Streptococcus mutans ATCC 25175 bacteria. The study used microtiter plate static biofilm assay. It used 2.08%, 4.17%, and 8.34% moringa leaf extract for the treatment group, 0.2% chlorhexidine gluconate for the positive control group, and aquadest for the negative control group. Microplate wells were filled with either 50 µL of extract, chlorhexidine, or aquadest, added with 40 µL of BHI, and then 10 µL of 1.5 x 108 CFU/ml bacterial suspension were inoculated. Each group was replicated 6 times. Microplate was incubated at 37oC for 24 hours, using microplate reader at 540 nm. Static analysis using ANOVA and Tukey HSD at p<0.05. One-way ANOVA results showed a significant difference on the inhibition percentage between groups, meaning that moringa leaf extract affected the adhesion ability of S. mutans ATCC 25175. The result of Tukey HSD post hoc test showed that moringa leaf extract with any concentration had a significant difference compared to 0.2% chlorhexidine gluconate in S. mutans ATCC 25175 inhibition. The conclusion of this study is that moringa leaf extract may inhibit bacterial adhesion of S. mutans ATCC 25175. The most effective concentration to inhibit S. mutans ATCC 25175 adhesion according to this study is 8.34%, even though it significantly is still below the potential of 0.2% chlorhexidine gluconate.
