SELEKSI INOKULUM DAN PENGATURAN pH UNTUK PEMISAHAN ASIDOGENESIS DAN METANOGENESIS PADA PROSES PRODUKSI BIOGAS DARI PALM OIL MILL EFFLUENT (POME)

Indonesia merupakan negara penghasil minyak kelapa sawit terbesar di dunia dengan total produksi pada tahun 2017 mencapai 34,47 juta ton. Tiap ton pengolahan Tandan Buah Segar (TBS) akan menghasilkan sekitar 2,4 hingga 3,7 ton limbah cair atau Palm Oil Mill Effluent (POME). POME merupakan limbah cair yang memiliki kandungan organik tinggi (sekitar 80.000 mg COD/L) yang sangat berpotensi menghasilkan biogas dengan proses anaerob. Salah satu jenis reaktor yang dapat digunakan untuk menguraikan POME menjadi biogas adalah two stage anaerobic digester. Penelitian ini dilakukan untuk mencari inokulum yang tepat untuk start-up pada masing-masing stage. Evaluasi dilakukan dengan metode microbial activity test. Microbial activity test bertujuan untuk mengetahui aktivitas bakteri dalam inokulum terhadap substrat ideal. Microbial activity test ini dilakukan untuk membandingkan kinerja 2 inokulum yaitu limbah biodiesel dan kotoran sapi yang telah diproses secara anaerob. Selain mengevaluasi kinerja inokulum, kondisi pH juga perlu dievaluasi untuk mengoptimalkan pemisahan proses asidogenesis dengan metanogenesis. Dua nilai pH yaitu pH 4 dan 5 dipilih untuk evaluasi kinerja bakteri pada reaktor asidogen agar proses metanogenesis terhambat dan pada pH 7 di reaktor metanogen agar proses asidogenesis terhambat. Khusus pada reaktor asidogen, injeksi udara dilakukan dalam jumlah kecil karena bakteri asidogen merupakan jenis bakteri fakultatif yang membutuhkan oksigen untuk mengoptimalkan degradasi substrat menjadi produk. Mula-mula penelitian dijalankan dengan substrat ideal, kemudian eksperimen yang sama dilakukan dengan POME sebagai substrat. Glukosa sebanyak 1,5 gr/L digunakan sebagai substrat ideal pada acidogenic activity test. Campuran asam asetat, asam propionat dan asam butirat dengan konsentrasi masing-masing 2:0,5:0,5 gr/L digunakan sebagai substrat ideal pada methanogenic activity test. Setelah tahap eksperimen substrat ideal, dilakukan pengujian lebih lanjut menggunakan substrat POME dengan variasi inokulum dan pH yang sama dengan substrat ideal. Perbandingan antara variasi proses ini dilakukan dengan membandingkan parameter kinetika dalam neraca massa substrat, volatile fatty acids (VFA), dan metana yang dihasilkan. Hasil penelitian acidogenic activity test dengan substrat ideal menunjukkan bahwa kinerja proses terbaik diperoleh pada reaktor dengan inokulum limbah anaerob biodiesel pH 4 yang dijalankan dengan injeksi udara, sedangkan methanogenic activity test substrat ideal menunjukkan bahwa kinerja proses terbaik juga diperoleh pada reaktor dengan inokulum limbah anaerob biodiesel. Sama halnya dengan eksperimen substrat ideal, proses asidogen terbaik pada substrat POME teramati pada reaktor dengan inokulum limbah anaerob biodiesel. Pada reaktor metanogen POME, performa reaktor terbaik diperoleh pada reaktor dengan inokulum limbah anaerob biodiesel dengan substrat yang berasal dari reaktor asidogen POME pada pH 4.

Indonesia is well-known as the world biggest palm oil producer with 34,47 million tons of crude palm oil (CPO) in 2017. Each ton of Fresh Fruit Bunches (FFB) will produce 2.4 to 3.7 tons of wastewater called Palm Oil Mill Effluent (POME). POME has high organic content (around 80,000 mg COD/L) which is potential to produce biogas by anaerobic process. One of the reactor types to process POME into biogas is two stage anaerobic digester. This research was conducted to find the best inoculum for the start-up of each stage in two stage anaerobic digester. Evaluation was conducted using microbial activity test method. Microbial activity test is a testing method to determine bacterial activity in the inoculum when it is grown in ideal substrate. This method was applied in this study to compare the performance of 2 inoculums i.e. anaerobically-digested biodiesel waste and anaerobically-digested cow manure. In addition to the performance evaluation of the inoculum, pH was also be evaluated to optimize the separation between acidogenesis process and metanogenesis process. Two pH values, i.e. pH 4 and 5 were chosen to evaluate the performance of bacteria in the acidogen reactor as well as to inhibit methanogenesis process. On the other hand, pH 7 was maintained in the methanogen reactor which was also aimed to inhibit the acidogenesis process. For acidogen reactors, micro-injection of air was introduced because acidogenic bacteria are facultative bacteria that need oxygen to optimize their performance. The experiments were first run with ideal substrate and followed with experiments using actual POME substrate. Aqueous glucose solution with the concentration of 1,5 gr/L was used as ideal substrate in acidogenic activity test, while the mixture of 2; 0,5; 0,5 gr/L for acetic, propionic, and butyric acids respectively was used as ideal substrate of methanogenic activity test. Microbial activity test on POME substrate was conducted with the aforementioned inoculums and the same pH as the ideal substrate. Comparison of those conditions was conducted by evaluating the kinetic parameters in the mass balances of substrate, volatile fatty acids (VFA), and methane. The results showed that the best reactor performance for ideal substrate in acidogenesis process was obtained at pH 4 with digested biodiesel waste as inoculum as well as with temporal micro-injection of air. Digested biodiesel waste also showed the best reactor performace for the ideal substrate in metanogenesis process. Similar to the ideal substrate experiment, the best acidogen reactor performance for POME substrate was the reactor with digested biodiesel inoculum. Meanwhile, biodiesel effluent reactor was also the best reactor for methanogen reactor with treated POME from the aforementioned acidogen reactors.
