IDENTIFIKASI MOLEKULER KUSKUS (Phalangeridae) ASAL MALUKU BERDASARKAN SEKUEN GEN PENYANDI NADH Dehidrogenase Sub-unit 1 (ND 1)

Kuskus merupakan mammalia berkantung (marsupial) dari famili Phalangeridae, yang sudah terkategori terancam punah (endangered) dan menuju kepunahan (vulnerable), oleh karena itu diperlukan kajian secara genetik sebagai dasar dalam klasifikasi kuskus untuk tujuan konservasi. Tujuan penelitian ini adalah untuk mengidentifikasi kuskus (Phalangeridae) asal Maluku secara molekuler berdasarkan sekuen gen penyandi NADH Dehidrogenase Sub-unit 1. Sampel DNA total diisolasi dari 14 kuskus asal Maluku. Amplifikasi segmen gen penyandi ND 1 menggunakan metode PCR dengan primer ND 1 Forward dan Primer ND 1 Reverse menghasilkan amplikon sepanjang 1275 bp. Pengolahan data menggunakan program MEGA versi 6.06 dengan penjajaran berganda program Clustal W menghasilkan sekuen nukleotida gen penyandi ND 1 sepanjang 957 nt. Hasil sekuensing gen ND 1 yang diperoleh selanjutnya dianalisis dan dibandingkan dengan Phalangeridae lain dari Genbank. Hasil analisis menunjukkan bahwa terdapat 161 situs nukleotida dan 25 situs asam amino yang dapat penjadi penanda genetik antara sampel kuskus dengan Phalanger vestitus dan Spilocuscus maculatus. Hasil identifikasi berdasarkan pohon filogenetik menggunakan metode Neighbor-Joining dengan nilai bootstrap 1000 kali menunjukkan bahwa sampel kuskus asal Maluku tediri dari Phalanger sp. dan S. maculatus.

Cuscus is a marsupial mammals (marsupial) of the family Phalangeridae, which has been categorized threatened (endangered) and toward extinction (vulnerable), therefore it is necessary to research genetically as a basis for classification cuscus for conservation purposes. The purpose of this research was to identify cuscus (Phalangeridae) from Maluku molecularly based sequences of genes encoding subunits of NADH dehydrogenase 1. Total DNA samples isolated from 14 cuscus from Maluku. Amplification of gene segments ND 1 using PCR with ND 1 Forward primers and ND 1 Reverse primers produce throughout 1275 bp amplicons. The data processing using the program MEGA version 6.06 with multiple alignment program Clustal W generate nucleotide sequences of ND 1 encoding genes along the 957 nt. ND 1 gene sequencing results were then analyzed and compared with other Phalangeridae from Genbank. The analysis showed that there were 161 sites of nucleotides and 25 sites of amino acid that may be genetic markers between the sample cuscus with P. vestitus and S. maculatus. Results of identification based phylogenetic trees using the Neighbor-Joining method with bootstrap value of 1000 times showed that cuscus samples from Maluku consist of Phalanger sp. and S. maculatus.
