EFEK ANTIFIBROTIK ISOLAT TAGITININ C DARI DAUN KEMBANG BULAN [Tithonia diversifolia (Hemsley) A. Gray] PADA FIBROBLAS KELOID

Latar Belakang: Keloid terjadi karena hiperproliferasi fibroblas dan peningkatan timbunan kolagen dengan karakteristik mirip dengan sel kanker. Tagitinin C merupakan senyawa golongan sesquiterpen lactones (SLs) yang diisolasi dari daun kembang bulan [Tithonia diversifolia (Hemsley) A. Gray]. Penelitian sebelumnya menyebutkan bahwa tagitinin C memiliki efek sitotoksik yang aktif dan selektif pada sel kanker kulit (M19) (IC50 = 0,996μg/mL; SI=40,536). Penelitian tentang efek antifibrotik tagitinin C terhadap fibroblas keloid belum pernah dilakukan. Tujuan: Mengkaji efek antifibrotik tagitinin C pada fibroblas keloid (FK) yang diukur dari proliferasi sel, timbunan kolagen FK, serta uji sitotoksik tagitinin C terhadap fibroblas normal (FN). Metode: Biakan lapis tunggal FK passase 3 diberi perlakuan tagitinin C dengan 8 serial dosis (0,0152) μg/mL selama 72jam dan 120jam. Kontrol positif menggunakan mitomisin C. Pengukuran viabilitas sel menggunakan metode MTT, sedangkan timbunan kolagen fibroblas keloid menggunakan metode sirius red. Pembacaan hasil dengan ELISA reader. Analisis data menggunakan regresi probit dengan program SPSS 19 for Windows. Hasil: Tagitinin C mampu menghambat viabilitas FK dengan IC50 0,122 μg/mL (inkubasi 72jam) dan 0,039 μg/mL (120jam), sedangkan mitomisin C IC50 0,120 μg/mL (72jam) dan IC50 0,100 μg/mL (120jam). Pada dosis IC50 tagitinin C memiliki timbunan kolagen fibroblas keloid 53,1% (72jam) dan 44,3% (120jam), sedangkan mitomisin C memiliki timbunan kolagen fibroblas keloid 60,4% (72jam) dan 52,1% (120jam). Pada dosis ≤ 2μg/mL tagitinin C tidak toksik pada fibroblast normal Kesimpulan: Kemampuan tagitinin C dalam menghambat viabilitas FK dan menurunkan timbunan kolagen keloid seirama dengan dosis (dose-dependent) dan waktu inkubasi (time-dependent). Tagitinin C memiliki tingkat toksisitas rendah pada FN.

Tagitinin C is a class of sesquiterpene lactones (SLS) were isolated from the leaves of the moon flower [Tithonia diversifolia (Hemsley) A. Gray]. Previous studies show that tagitinin C has a cytotoxic effect on active and selective skin cancer cell (M19) (IC50 = 0,996μg / mL; SI = 40,536). Research on the effects of antifibrotik tagitinin C on keloid fibroblasts has not been done. Objective: Evaluate the effects of tagitinin C from T.diversifolia to monolayer keloid fibroblasts (KF) has on cell proliferation, collagen deposition, and cytoxicity on normal fibroblast (NF). Methods: Monolayer cultures of more than three passages of keloid fibroblast were exposed with 8 serial doses of tagitinin C (0.015 to 2) μg / mL during 72 and 120 hour. Positive control using mitomycin C. Cellular viabilities were measured by MTT assay. Collagen depositions were measured by Sirius Red assay for nonsoluble collagen. The reading of the results of the ELISA reader. Data were analyzed by regression probit with SPSS 19 for Windows. Results: Tagitinin C can inhibit KF viability with IC50 0.122 μg/mL (72h) and 0.039 μg/mL (120h), whereas mitomycin C IC50 0.120 μg/mL (72h) and IC50 of 0.100 μg/mL (120h). At IC50 dose of Tagitinin C on keloid collagen deposition 53.1% (72h) and 44.3% (120h), whereas the IC50 dose of mitomycin C on keloid collagen deposition 60.4% (72h) and 52.1% (120h) . At doses ≤ 2μg/mL tagitinin C is not toxic on normal fibroblast Conclusion: The ability of tagitinin C inhibit viability KF and lowering keloid collagen deposition are consistent with the dose (dosedependent) and incubation time (timedependent). Tagitinin C has a low toxicity level on NF.
