Kadar testosteron serum, spermatogenesis, dan kualitas spermatozoa, mencit jantan galur BALB/c setelah diberi ekstrak daun senggani (Melastoma polyanthum BI)

Senggani (Melastoma polyanthum BI.) merupakan salah satu tanaman obat, daun senggani telah digunakan sebagai obat kontrasepsi secara tradisional. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun senggani (Melastoma polyanthum BI.) terhadap kadar testosteron serum, spermatogenesis, dan kualitas spermatozoa mencit jantan (Mus musculus L.) serta waktu pemulihannya. Duapuluh empat ekor mencit jantan galur Balb/c berumur 8 minggu. Sampel penelitian dibagi secara acak menjadi 2 kelompok dan masing – masing kelompok dibagi menjadi 4 sub kelompok perlakuan dosis yaitu A= kontrol ( 0 mg/20g BB), B = Dosis 1 (35 mg/20g BB), C = Dosis 2 (70 mg/20g BB) dan D = Dosis 3 (105 mg/20g BB). Tiap kelompok terdiri atas 3 ekor hewan uji sebagai ulangan. Perlakuan diberikan secara oral, sebanyak 0,5ml/hari selama 35 hari. Setelah 35 hari dilakukan pengamatan terhadap kelompok I (12 ekor hewan uji dikorbankan) sedangkan pengamatan II dilakukan 35 hari setelah perlakuan dihentikan (12 ekor hewan uji dikorbankan), untuk pengamatan pemulihannya. Pengamatan dilakukan terhadap kadar hormon testosteron serum dengan metode RIA (Radio Immuno-assay). Untuk menghitung lapisan sel spermatogenesis dibuat preparat histologi testis dengan metode paraffin, pewarnaan Hematoksilin-Eosin (HE), dan analisis kualitas spermatozoa mencit dengan Neubauer. Data yang diperoleh diuji dengan Analysis of Variance Test (ANOVA a = 0,05) dan Duncan´s Multiple Range Test (DMRT) dilanjutkan dengan regresi linier. Berdasarkan pengamatan dan analisis statistik didapatkan hasil bahwa perlakuan dengan ekstrak daun senggani dengan dosis yang berbeda berpengaruh nyata terhadap semua parameter yang diamati. Hal ini ter lihat jelas dengan semakin besar dosis yang diberikan maka terjadi penurunan kadar testosteron serum, jumlah sel spermatogenik dan kualitas spermatozoa mencit. Pengaruh ekstrak daun senggani bersifat sementara karena dapat pulih kembali hal ini dapat dilihat dengan terjadinya peningkatan kadar hormon tetosteron, spermatogenesis dan kulitas spermatozoa mencit setelah 35 hari perlakuan ekstrak daun senggani dihentikan.

Senggani (Melastoma polyanthum BI.) is categorized into herbs, which has been used as a traditional contraceptive, particularly its leaves. The research aims to study the effects of Melastoma polyanthum BI leaves extract on serum testosterone level, spermatogenesis, and spermatozoa quality of male mice (Mus musculus L.) as well as its recovery period. Twenty four male mice aged 8 week, Balb/c. From 24 mice were divided randomizely into 2 groups, each group into 4 sub groups of dose treatment, i.e. A = control (0mg/gram body weight ( gbw), B = Dose -1 (35 mg / gbw), C = Dose - 2 (70 mg /gbw), D = Dose-3 (105 mg/ gbw). Each subgroup of treatment was provided with three mice for replication. A 0.5 ml/day of senggani extract was administered orally to mice for 35 days. On day 35, an observation on serum testosterone level was done by using RIA (Radio Immuno-assay) method. The testes histological preparation were made using paraffin satained with Hematoksilin- Eosin (HE) method to calculate the number of spermatogenic cell layers, and analysis spermatozoa quality by using Neubauer, was performed on group-1 (where 12 mice were killed), while II was done 35 days later (where 12 mice were killed) for recovery observation. The collected data were tested using Analysis of Variance Test (ANOVA a = 0.05) followed by a testing using Duncan´s Multiple Range Test (DMRT) and Linear regression analysis. Based on the observation and statistical analysis, the results show that different dose treatment of senggani leave extract affects all the tested parameters significantly. The higher the dose given, the lower the testosterone level, spermatogenetic cell quantity, and spermatozoa quality decrease. The effect of senggani is temporary which means that mice could recover from senggani effects. It can be observed through the increase in testosterone level, spermatogenesis, and spermatozoa quality of mice after 35 days after senggani extract treatment was stopped.
