PENGARUH PENAMBAHAN INOKULUM Lactobacillus plantarum HL-15 DAN VARIASI KOTAK FERMENTASI TERHADAP KARAKTERISTIK KIMIA DAN MIKROBIOLOGI COKELAT SUSU

Kakao (Theobroma cacao L.) merupakan salah satu komoditas penting di Indonesia. Salah satu olahan biji kakao yang paling populer adalah cokelat susu (milk chocolate). Kualitas akhir cokelat susu seperti ketahanannya terhadap jamur dapat dipengaruhi oleh proses pengolahannya. Salah satu permasalahan selama proses pengolahan cokelat Indonesia adalah munculnya jamur penghasil toxin pada biji kering kakao. Kontaminasi jamur penghasil mikotoksin dapat berasal dari kotak fermentasi dari kayu yang berulang digunakan. Proses fermentasi tradisional dapat dikendalikan salah satunya dengan penambahan starter bakteri asam asam laktat yang memproduksi metabolit senyawa antijamur. Tujuan penelitian ini adalah untuk mengetahui pengaruh variasi kotak fermentasi lama dan baru serta penambahan starter Lactobacilus plantarum HL-15 yang memiliki aktivitas antijamur pada proses fermetasi terhadap kualitas kimia dan mikrobiologis cokelat susu. Penambahan kultur starter Lactobacilus plantarum HL-15 pada saat fermentasi tidak mempengaruhi pH cokelat namun dapat memberikan konsentrasi cemaran jamur dan yeast 1 log cycle lebih rendah (6,5 x 102 koloni/gram dan 4,5 x 102 koloni/gram) daripada biji fermentasi tanpa starter (2,55 x 103 koloni/gram dan 1,65 x 103 koloni/gram). Variasi penggunaan kotak fermentasi lama dan baru tidak mempengaruhi pH cokelat namun penggunaan kotak fermentasi baru memberikan hasil kosentrasi jamur dan yeast yang lebih rendah pada media DG-18 dan inkubasi 5 hari di suhu ruang dibandingkan dengan kotak lama. Cokelat susu dengan kualitas mikrobiologis terbaik ditunjukkan dengan jumlah cemaran jamur paling kecil adalah cokelat susu yang pada saat fermentasi biji digunakan kotak baru dan dilakukan penambahan kultur starter Lactobacilus plantarum HL-15 (4,5 x 102 koloni/gram).

Cocoa (Theobroma cacao L.) is one of the most important comodity in Indonesia. One of the most popular products of cocoa is milk chocolate. The chemical and microbiological quality of milk chocolate can be affected by it’s production process. One problem which appears during dry cocoa beans process is the contamination of toxigenic mold. The contamination of the toxigenic mold can come from the usage of old fermentation box. The fermentation prosess can be controlled by the addition of starter such as lactic acid bacteria which can produce antifungal metabolites. The objective of this research is to observe the effect of the usage of fermentation box and the Lactobacilus plantarum HL-15 culture starter addition to the chemical dan microbiological quality of milk chocolate. The Lactobacilus plantarum HL-15 culture starter addition in the fermentation prosess does not affect the pH of milk chocolate, in the other hand it can give lower concentration of mold and yeast in the final products (6,5 x 102 colony/gram and 4,5 x 102 colony/gram) compared with the chocolate without starter addition (2,55 x 103 colony/gram and 1,65 x 103 colony/gram). The variation of fermentation box also does not gives a significant different to the pH of milk chocolate, but the usage of new fermentation box gives lower concentration of mold and yeast in the final products (4,5 x 102 colony/gram and 1,65 x 103 colony/gram) compared with the chocolate without starter addition (6,5 x 102 colony/gram and 2,55 x 103 colony/gram) in DG-18 media. Milk chocolate with the best microbial quality shown by the lowest concentration of mold and yeast contamination is observed in the addition of starter Lactobacilus plantarum HL-15 and the usage of new fermentation box (4,5 x 102 colony/gram).
