AKTIVITAS ANTIVIRUS FRAKSI ETIL ASETAT Streptomyces sp. GMR22 TERHADAP VIRUS DENGUE SEROTIPE 1

Virus dengue serotipe 1 merupakan serotipe yang dominan di beberapa wilayah Indonesia dan terdapat hampir di semua daerah endemik dengue sehingga perlu mendapat perhatian. Senyawa antivirus yang efektif untuk virus dengue belum ditemukan. Streptomyces sp. GMR22 diisolasi dari tegakan tanaman kayu putih di Hutan Wanagama I Yogyakarta. Isolat ini diduga memiliki aktivitas antivirus terhadap virus dengue serotipe 1. Penelitian sebelumnya menunjukkan bahwa ekstrak etil asetat Streptomyces sp. GMR22 sangat toksik terhadap sel BHK-21 yang merupakan sel uji dalam uji antivirus dengue. Penelitian ini bertujuan untuk mengetahui sitotoksisitas fraksi etil asetat Streptomyces sp. GMR22 terhadap sel BHK-21 dan aktivitas antivirusnya terhadap virus dengue serotipe 1. Penelitian ini diawali dengan kultur dan fermentasi Streptomyces sp. GMR22, ekstraksi bertingkat dengan n-heksan dan etil asetat, fraksinasi dengan kromatografi kolom, uji sitotoksik dengan WST-1 assay, propagasi virus, penghitungan titer virus dengan plaque assay, uji antivirus dengan qRT-PCR dan pengamatan viabilitas sel. Proses fraksinasi menghasilkan 8 subfraksi gabungan. Nilai CC50 subfraksi gabungan 1,2,3,4,6,7,8, dan fraksi etil asetat adalah >200 mikro gram/mili liter sedangkan subfraksi gabungan 5 adalah 190,98 mikro gram/mili liter. Titer virus adalah 2,125x105 PFU/ml. Uji antivirus menunjukkan bahwa penghambatan virus paling baik dimiliki oleh subfraksi gabungan 4 (kloroform:methanol, 95:5). Pada dosis 5, 50, 100 dan 200 mikro gram/mili liter, subfraksi gabungan 4 dapat menghambat perkembangan virus dengue serotipe 1 tanpa menyebabkan kerusakan sel BHK-21 yang berarti, dibandingkan dengan subfraksi yang lain.

Dengue virus serotype 1 was the predominant dengue serotype in some region of Indonesia and there was almost in all endemic areas of dengue and should get attention. Effective dengue antiviral compounds has not been found. Streptomyces sp. GMR22 was isolated from the stand of eucalyptus plants in Wanagama I Forest Yogyakarta. This isolate thought to have antiviral activity against dengue virus serotype 1. Previous studies showed that the ethyl acetate extract of Streptomyces sp. GMR22 was toxic to BHK-21 cell which was the host cells in dengue antiviral study. Objective of this study was to determine how was cytotoxicity of ethyl acetate fraction of Streptomyces sp. GMR22 to the BHK-21 cell and its antiviral activity against dengue virus serotype 1. This study was started by culture and fermentation of Streptomyces sp. GMR22, gradient extraction with n-hexane and ethyl acetat, fractionation using column chromatography, WST-1 assay to get fraction CC50 value, virus propagation, virus titration with plaque assay, antiviral test using qRT-PCR and cell viability study. Fractionation process resulted in 8 combined subfractions. CC50 value of combined subfraction 1,2,3,4,6,7,8, and ethyl acetate fraction was >200 micro grams/mili litres whereas combined subfraction 5 was 190,98 micro grams/mili litres. Virus titer was 2,125x105 PFU/ml. Antiviral test show that combined subfraction 4 (chloroform:methanol, 95:5) inhibited virus growth better than others. Dosage 5,50,100, and 200 micro grams/mili litres of combined subfraction 4 inhibited virus growth without causing significant BHK-21 cell damage, compared to other subfraction.
