AKTIVITAS ANTIVIRUS FRAKSI EKSTRAK ETIL ASETAT METABOLIT SEKUNDER Streptomyces sp. GMY01 TERHADAP VIRUS DENGUE SEROTIPE-1

Demam berdarah merupakan salah satu penyakit yang dapat menimbulkan gejala klinis serius hingga menyebabkan kematian, sehingga terapi terhadap penderita demam berdarah perlu dioptimalkan. Sampai saat ini belum ada obat antiviral yang efektif digunakan untuk terapi pasien demam berdarah. Ekstrak etil asetat dari metabolit sekunder Streptomyces sp. GMY01 diketahui memiliki efek antiviral terhadap virus DEN-1. Namun, belum diketahui senyawa aktif yang bertanggungjawab terhadap aktivitas tersebut. Penelitian ini dilakukan untuk memisahkan komponen dalam ekstrak etil asetat metabolit sekunder Streptomyces sp. GMY01 dengan fraksinasi dan kemudian menguji potensi antiviral dari masing-masing fraksi untuk mengetahui keberadaan senyawa aktif antiviral terhadap virus DEN-1. Metabolit sekunder Streptomyces sp. GMY01 didapatkan dengan fermentasi isolat Streptomyces sp. GMY01 dalam media SNB selama 15 hari. Metabolit sekunder yang dihasilkan kemudian diekstraksi menggunakan etil asetat dengan metode partisi dan dilanjutkan dengan fraksinasi menggunakan metode kromatografi kolom. Uji WST-1 assay dilakukan untuk mengetahui CC50 masing-masing fraksi untuk memprediksi dosis yang akan diberikan dalam uji antiviral terhadap virus DEN-1. Uji antiviral dilakukan dengan metode semi kuantitatif RT-PCR. Kemampuan penghambatan virus DEN-1 dianalisis dengan hasil kuantifikasi band RT-PCR menggunakan program Image-J. Hasil penelitian menunjukkan bahwa fraksi gabungan 6 memiliki persentase penghambatan pertumbuhan virus terbesar dibandingkan dengan fraksi gabungan lain, yaitu sebesar 70%. Persentase penghambatan ini lebih tinggi apabila dibandingkan dengan persentase penghambatan pertumbuhan virus dari ekstrak etil asetat dan supernatan metabolit sekunder Streptomyces sp. GMY01.

Dengue fever is one kind of a disease that can lead into serious clinical symptoms and cause death. Clinical therapy for patiens with dengue fever should be optimized to reduce fatality number caused by this disease. To date, there has been no approved antiviral drug used to treat dengue patients so the research on antiviral of dengue virus is very important to do and developed. Antiviral activity against DEN-1 virus of ethyl acetat extract from secondary metabolites Streptomyces sp. GMY01 has been reported. But, the active compound that responsible for that antiviral activity is still unknown. In this study, in vitro experiment was conducted to separate all active compounds in the ethyl acetate extract into fractions and then identificate which fraction containing the active antiviral compound. Fermentation of Streptomyces sp. GMY01 was carry out for 15 days to obtain the secondary metabolites. Active compound was extracted from supernatant using partition method and ethyl acetate as extraction solvent and followed by fractionation using column chromatography method. WST-1 assay was conducted to analyze effective dose that can be used for antiviral experiment with the help of CC50 value. In this experiment, semi-quantitative RT-PCR had been used to determine the antiviral activity and IMAGE-J as quantification software. This study showed that the highest antiviral percentage, 70%, was obtained from the sixth fraction. These percentage was also higher than the antiviral percentage of crude extract and crude supernatant.
