ISOLASI DAN IDENTIFIKASI SENYAWABERPOTENSI ANTITUMOR DARI KAPANGYANG BERASOSIASI DENGAN ORGANISME LAUT

Kapang yang berasal dari laut merupakan sumber senyawa aktif yang dapat dikembangkan sebagai bahan obat untuk industri farmasi. Penelitian ini bertujuan untuk: 1) melakukan penapisan kapang laut penghasil senyawa sitotoksik dari beberapa wilayah perairan di Indonesia; 2) mengisolasi dan mengidentifikasi senyawa antikanker yang dihasilkan; 3) mengetahui efek sitotoksik, pengaruh senyawa aktif terhadap siklus sel T47D dan kemampuan induksi apoptosis senyawa aktif yang dihasilkan. Kapang diisolasi dari organisme laut yang diambil dari beberapa perairan di Indonesia yaitu Taman Nasional Laut Wakatobi-Sulawesi Tenggara, Pantai Binuangeun-Banten, perairan sekitar kota Manado-Sulawesi Utara, dan Taman Nasional Laut Kepulauan Seribu-Jakarta. Isolasi kapang dilakukan dengan menggunakan media Malt Extract Agar (MEA), Minimum Fungi Medium (MFM) and Glucose Peptone Yeast medium (GPY). Kapang diidentifikasi berdasarkan ciri-ciri morfologi dan molekular. Strain aktif dikultivasi pada labu 3L sebanyak 20 flask, masing-masing mengandung 1 L media SWS (mengandung 1% pati dapat larut, 0,2% pepton soya, 1 L air laut) dan diinkubasi dalam kondisi statis pada suhu 27-28oC selama 5 minggu. Miselium kapang diekstraksi dengan campuran pelarut diklorometan-metanol (1 : 1 v/v) sedangkan broth diekstraksi dengan menggunakan etil asetat. Fraksinasi dilakukan dengan menggunakan kolom vakum SiO2 2,5 x 40 cm dan isolasi senyawa aktif dilakukan dengan menggunakan kromatografi lapis tipis preparatif. Senyawa aktif dielusidasi dengan menggunakan data spektroskopik UV, FT-IR, LC-ESI-Tof-MS, FT-NMR (1H-NMR, 13C-NMR, DEPT dan HMQC). Efek sitotoksik terhadap sel T47D, HepG2, HeLa dan Vero diuji dengan metode MTT. Analisis siklus sel T47D dilakukan dengan flow cytometri dengan menggunakan senyawa fluoresen propidium iodida. Analisis flow cytometry juga digunakan untuk mengetahui efek senyawa aktif dalam menginduksi apoptosis pada sel T47D dengan menggunakan senyawa fluoresen Annexin-V-FLUOS. Ekstrak kapang yang dihasilkan (45 ekstrak) telah diuji aktivitasnya terhadap sel T47D pada konsentrasi 30 μg/ml, dari ke-45 ekstrak kapang tersebut, ekstrak dari kapang MFW39 menunjukkan aktivitas sitotoksik yang paling kuat. Kapang ini diisolasi dari ascidia Aplidium longithorax yang diperoleh dari Taman Nasional Laut Wakatobi. Berdasarkan identifikasi secara morfologi dan molekular, kapang MFW39 termasuk dalam jenis Emericella nidulans. Isolasi kapang E.nidulans dari ascidia A.longithorax baru pertama kali ini dilaporkan. Senyawa sitotoksik yang terdapat dalam kapang MFW39 berhasil diisolasi dari ekstrak miselium. Berdasarkan spektra LC-ESI-ToF-MS, senyawa tersebut memiliki rumus molekul C27H22N2O10S2 (m/z 597.1105, [M-H]¯). Elusidasi struktur molekul berdasarkan spektra UV, FT-IR, LC-ESI-ToF-MS, 1H-NMR, 13C-NMR, DEPT dan HMQC menunjukkan bahwa senyawa tersebut adalah emestrin. Emestrin memiliki aktivitas sitotoksik terhadap sel T47D, HepG2, HeLa dan Vero dengan nilai IC50 berturut-turut sebesar 1,7; 4,3; 12,8 dan 258,8 μg/ml. Berdasarkan analisis siklus sel dengan menggunakan sel T47D, perlakuan emestrin pada konsentrasi 1,0 μg/ml menyebabkan cell-cycle arrest pada fase G0/G1, sedangkan pada konsentrasi 3,0 μg/ml menyebabkan munculnya sub-populasi sel G1 pada kromatogram. Analisis flow cytometry dengan menggunakan Annexin-V-FLUOS memperlihatkan bahwa hampir sebagian besar sel T47D yang diberi perlakuan emestrin pada dosis 1,0 dan 3,0 μg/ml mengalami apoptosis (masing-masing sebesar 83,6% dan 92,6%). Efek emestrin terhadap siklus sel dan induksi apoptosis pada sel T47D belum pernah dilaporkan sebelumnya. Berdasarkan uji sitotoksik, analisis siklus sel dan uji induksi apoptosis, dapat disimpulkan bahwa emestrin berpotensi untuk diteliti lebih lanjut aktivitasnya sebagai senyawa antikanker.

Marine-derived fungi have proven to be a rich source of structurally unique and biologically active secondary metabolites for the development of new pharmaceutical agents. This research was aimed to: 1) screen cytotoxic activity of marine fungi collected from several Indonesian waters; 2) isolate and identify active compound from marine fungi; 3) determine cytotoxicity potential, cell cycle analysis and apoptosis effect of active compound. The fungi were isolated from marine organism collected from Wakatobi Marine National Park, South East Sulawesi, Binuangeun Beach, Banten, Manado waters, North Sulawesi and Kepulauan Seribu Marine National Park, Jakarta. Isolation of fungi was conducted by using Malt Extract Agar (MEA), Minimum Fungi Medium (MFM) and Glucose Peptone Yeast medium (GPY). Molecular identification of fungus was determined based on morphology and molecular characteristic. The active strain was cultivated in a flask (3 L) for 20 flasks, containing 1 L of SWS medium (containing soluble starch 1%, soya peptone 0.2% and 1 L sea water), each flask incubated in static condition at 27-28oC for 5 weeks. Mycelium was extracted with mixture of dichloromethane-methanol (1 : 1 v/v) whereas broth was extracted by using ethyl acetate. Fractionation was performed by using 2,5 x 40 cm SiO2 vacuum column chromatography and purification was done by SiO2 preparative thin layer chromatography. The active compound was elucidated with spectroscopic analysis by using FT-IR, LC-ESI-ToF-MS, FT-NMR (1H-NMR, 13C-NMR, DEPT and HMQC). Cytotoxic effect of active compound against T47D, HepG2, HeLa and Vero cells were assayed using MTT test. Cell cycle distribution of T47D cells exposed to the active compound was investigated by using flow cytometry and the ability of this compound to induce apoptosis of T47D cells was examined by Annexin-V-FLUOS using flow cytometry. Marine fungi extract (45 extracts) were tested against the growth of T47D cell at extract concentration of 30 μg/ml, among them, strain of MFW39 exhibited the strongest cytotoxic activity. This strain was isolated from ascidia Aplidium longithorax collected from Wakatobi Marine National Park. The MFW39 marine fungal was identified as Emericella nidulans based on its morphology and molecular features. Isolation of this fungal from A.longithorax ascidan was first reported so far. The cytotoxic compound was isolated successfully from mycelium extract of MFW39 fungal. Based on LC-ESI-ToF-MS spectra, the molecular formula of this compound was established as C27H22N2O10S2 (m/z 597.1105, [M-H]¯). Elucidation of molecular structure using spectra data of FT-IR, LC-ESI-ToF-MS, 1H-NMR, 13C-NMR, DEPT and HMQC techniques concluded that the active compound was emestrin. Emestrin was cytotoxic against T47D, HepG2, Hela and Vero cells having IC50 values of 1.7; 4.3; 12.8 and 258,8 μg/ml, respectively. Based on the cell cycle analysis, treatment at with emestrin (1.0 μg/ml) was able to induce cell-cycle arrest in G0/G1 phase, and when the concentration was tripled (3.0 μg/ml), a sub-population of cells (sub G1) appeared. The flow cytometry analysis using Annexin-V-FLUOS revealed that most of T47D cell treated with the emestrin at 1.0 and 3.0 μg/ml undergone apoptosis (83.6% and 92.6 %, respectively). Effect of emestrin to T47D cell cycle and its apoptosis induction had not been reported yet. Based on the cytotoxicity test, cell cycle analysis and apoptosis assay, we can infer that the emestrin was potential to be developed as anticancer agent.
