AKTIVITAS SITOTOKSIK EKSTRAK ETANOLIK DAN FRAKSI ETIL ASETAT DARI DAUN SAMBUNG NYAWA (Gynura procumbens (Lour.) Merr) SERTA KOMBINASINYA DENGAN 5-FLUOROURASIL PADA SEL CT 26

Strategi kombinasi kemoterapi (ko-kemoterapi) dibutuhkan sebagai agen terapi kanker melalui kombinasi suatu senyawa dengan agen kemoterapi. Dengan mengetahui potensi sitotoksik ekstrak etanolik dan fraksi etil asetat daun sambung nyawa atau Gynura procumbens (Lour.) Merr pada sel CT 26, dapat dikembangkan agen sitotoksik kombinasi senyawa tersebut dengan 5-Fluorourasil (5-FU). Dari ko-kemoterapi tersebut diharapkan dapat memberikan efek sitotoksik yang sinergis dan mampu menurunkan viabilitas lebih tinggi daripada perlakuan tunggal senyawa pada sel kanker kolon CT 26. Bahan uji dalam penelitian ini meliputi ekstrak etanolik dan fraksi etil asetat dari daun sambung nyawa atau Gynura procumbens (Lour.) Merr (EE dan FEA GP), serta 5-FU. Analisis kandungan senyawa dilakukan dengan metode Kromatografi Lapis Tipis (KLT). Uji sitotoksisitas tunggal dan kombinasi dilakukan selama 24 jam menggunakan MTT assay dengan nilai IC50 dan CI sebagai parameter sitotoksisitas dan sinergisitas kombinasi. Profil KLT FEA dan EE GP belum menunjukkan adanya kandungan senyawa rutin maupun kuersetin. Berdasar hasil uji sitotoksik tunggal, EE dan FEA GP masing-masing menunjukkan aktivitas sitotoksik dengan memberikan nilai IC50 sebesar 1772 dan 468 �µg/mL, sedangkan 5-FU memberikan nilai IC50 sebesar 11 �µg/mL. Kombinasi EE dan FEA GP masing-masing dengan 5-FU memiliki efek sitotoksik yang sinergis dan memberikan aktivitas penurunan viabilitas terhadap sel CT 26 lebih tinggi dibanding perlakuan tunggalnya hanya pada kombinasi dosis tertentu.

Combination of chemotherapy (co-chemotherapy) is needed as a cancer therapeutic agent through a combination of a chemical compound with a chemotherapy agent. Cytotoxic activity of ethanolic extract and ethyl acetate fraction of Gynura procumbens (Lour.) Merr leaves on colorectal cancer cell line (CT 26 cell) can be developed as co-chemotherapy agent in combination with 5-Fluorouracil (5-FU). This study aimed to assess whether co-chemotherapy produced synergistic effect and decrease the cell viability higher than the single treatment on CT 26 colon cancer cells. Material were used in this study was ethanolic extract and ethyl acetate fraction of Gynura procumbens (Lour.) Merr leaves (EE and FEA GP), also 5-FU. The chromatography profile of EE and FEA GP was determined using Thin Layer Chromatography (TLC). MTT assay was performed to determine the growth inhibitory effect of both single (EE GP, FEA GP or 5-FU) and combination treatments on the growth of CT 26 for 24 hours. There was no rutin and quercetin identified from TLC profile of EE and FEA GP. EE GP, FEA GP, 5-FU inhibited cell growth in a dose dependent manner and exhibited an IC50 value of 1772, 468 and 11 �µg/mL, respectively. When EE and FEA GP was combined with 5-FU, it caused synergism and decrease the CT 26 cell viability higher than the single treatment, but it happened only on certain doses.
