DETEKSI PENINGKATAN AKTIVITAS ENZIM ESTERASE (Est) NON SPESIFIK PADA NYAMUK Aedes aegypti, L (DIPTERA: CULICIDAE) DI SEKIP DAN POGUNG KIDUL

Latar Belakang. Dengue masih menjadi penyakit menular yang serius di Indonesia. Ratusan hingga ribuan kasus baru terjadi setiap tahunnya. Fakultas Kedokteran Universitas Gadjah Mada dan daerah sekitarnya (Sekip dan Pogung Kidul) merupakan daerah padat penduduk dan merupakan tempat yang disenangi nyamuk Ae. aegypti. Penggunaan insetisida pun tak lepas dari daerah tersebut. Penggunaan yang ekstensif dan tidak tepat telah membuat munculnya resistensi terhadap insektisida pada nyamuk Ae. aegypti. Spesies ini mengalami berbagai jenis resistensi pada berbagai jenis insektisida. Insektisida yang paling sering digunakan antara lain sipermetrin dan malation. Salah satu mekanisme resistensi yang penting adalah peningkatan aktivitas enzim detoksifikasi seperti cytochrome 450 monooxygenase, esterase dan gluthatione-S-transferase, tergantung pada jenis serangga atau galur dan juga jenis insektisida. Aktivitas enzim esterase non spesifik pada nyamuk Ae. aegypti perlu diteliti untuk mengetahui status kerentanan dan mekanisme resistensi nyamuk di daerah tersebut. Tujuan. Menentukan aktivitas enzim esterase non spesifik dan status resistensi nyamuk Ae. aegypti yang berasal dari daerah Sekip dan Pogung Kidul. Metode. Koleksi larva dan pupa dilakukan di wilayah Sekip dan Pogung Kidul yaitu di RT 01, 02 dan 04. Larva lalu dikolonisasi di Lab Parasitologi FK UGM. Aktivitas enzim esterase non spesifik diukur dengan metode Lee (1992) dengan substrat α-naftil asetat. Hasil. Rerata nilai AV nyamuk Ae aegypti dari Sekip adalah 0,264 ± 0,080 sedangkan Pogung Kidul 0,231 ± 0,057. Nilai cut off positive sebesar 0,339. Kesimpulan. Sebagian besar sampel (87,5%) dari Sekip dan 93,75% dari Pogung Kidul memiliki nilai AV lebih rendah dari nilai cut off positive. Nyamuk Ae. aegypti yang berasal dari Sekip dan Pogung Kidul masih rentan, namun berpotensi menjadi resisten terhadap insektisida yang berkaitan dengan aktivitas enzim esterase non spesifik yang mampu menghidrolisis alfa naftil asetat.

Background. Dengue is still one of the most important infectious disease in Indonesia. Hundreds to thousands of new cases emerge annually Faculty of Medicine Universitas Gadjah Mada and its surrounding areas( Sekip and Pogung Kidul) are housing complexes with high density population and also the important habitat and breeding area of Ae. aegypti. The use of insecticides was done by the people of that area. Extensive and inappropriate use has made the emergence of Ae. aegypti resistance. This species has undergone some resistances against some insecticides. The most common used insecticides are cypermethrin and malathion. One of the resistance mechanism is increase in detoxifying enzymes such as cytochrome 450 monooxygenase, esterase and gluthatione-S-transferase. Non specific esterase activity needs to be measured so the susceptibility status and resistance mechanism can be known. Aim. To determine non specific esterase activity and the resistance status of Ae. aegypti from Sekip and Pogung Kidul, Sleman, Yogyakarta. Method. Larvae and pupae were collected at Sekip and RT 01, 02 and 04 of Pogung Kidul. Larvae then were colonized in Parasitology Laboratorium of FK UGM. Non specific esterase activity was measured with Lee method (1992) with α-naphtyl acetate as substrate. Result. Mean AV of Ae aegypti from Sekip was 0,264 ± 0,080 and from Pogung was 0,231 ± 0,057. The value of cut off positive was 0,339. Conclusion. Most sample (87,5%) from Sekip and 93,75% from Pogung Kidul had the AV value below cut off positive. The mosquitoes from Sekip and Pogung Kidul were sensitive but had the potential to become resistant against insecticides associated with non specific esterase activity which capable of hydrolising alpha naphtyl acetate.
