UJI AKTIVITAS ANTIBAKTERI EKSTRAK ETANOL DAUN POHPOHAN (Pilea melastomoides Wedd) TEHADAP BAKTERI Porphyromonas gingivalis

Bakteri Porphyromonas gingivalis merupakan bakteri anaerob Gram negatif yang terdapat di dalam mulut dan diduga merupakan salah satu penyebab utama bau mulut karena kemampuannya menghasilkan VSCs (Volatile Sulfure Compounds). Daun pohpohan (Pilea melastomoides Wedd) merupakan lalapan yang sering dikonsumsi masyarakat Jawa Barat yang mengandung senyawa flavonoid, alkaloid, steroid dan telah diteliti mempunyai aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak etanol daun pohpohan terhadap P. gingivalis, serta golongan senyawa aktif yang memiliki aktivitas antibakteri. Pohpohan diekstraksi menggunakan etanol 70% dengan cara maserasi. Uji aktivitas antibakteri dilakukan dengan metode mikrodilusi untuk mengetahui nilai Konsentrasi Hambat Minimum. Setiap sumuran pada mikroplate diisi dengan ekstrak pohpohan yang dilarutkan dengan DMSO 10% dengan konsentrasi akhir 1000 ���µg/mL; 500 ���µg/mL; 250 ���µg/mL;125 ���µg/mL; 62,5 ���µg/mL; 31,25 ���µg/mL dan kontrol positif klorheksidin 1%. Penghambatan pertumbuhan bakteri diukur dengan microplate reader setelah dilakukan penambahan MTT pada tiap sumuran. Hasil uji berupa data optical density yang digunakan untuk menghitung nilai % penghambatan. Nilai IC50 ditentukan dengan metode probit dengan menggunakan program SPSS versi 24. Deteksi senyawa aktif yang memiliki aktivitas sebagai antibakteri dilakukan dengan bioautografi kontak dengan fase diam silika gel F254 dan fase gerak n-heksan : etil asetat (1:1). Hasil penelitian menunjukkan nilai KHM50 adalah 214,98 ���µg/mL sedangkan KHM90 pada konsentrasi 2943,37 ���µg/mL. Hasil bioautografi menunjukkan zona jernih pada hRf 75-96. Golongan senyawa aktif antimikroba kemungkinan adalah flavonoid dan terpenoid.

Porphyromonas gingivalis is an anaerobic Gram negative bacteria found in the mouth and is thought to be one of the main causes of bad breath due to its ability to produce VSCs (Volatile Sulfure Compounds). Pohpohan leaves (Pilea melastomoides Wedd) is a vegetable that is often consumed by the people of West Java that containing flavonoids, alkaloids, steroids and has been studied to have antibacterial activity. This study aims to determine the antibacterial activity of extract ethanol from pohpohan leaf to P. gingivalis, as well as classes of active compounds that have antibacterial activity. Pohpohan was extracted using 70% ethanol by maceration. The antibacterial activity test was performed by microdilution method to find out the value of Minimum Inhibition Concentration. Each well on microplate was filled with pohpohan extract dissolved with DMSO 10% with final concentration 1000 ���µg/mL; 500 ���µg/mL; 250 ���µg / mL; 125 ���µg/mL; 62,5 ���µg/mL; 31.25 ���µg/mL and positive control of chlorhexidine 1%. Inhibition of bacterial growth was measured by microplate reader after the addition of MTT at each well. The test result is optical density data which is used to calculate the % inhibition value. IC50 values were determined by probit method using SPSS version 24 program. Detection of active compound that have activity as antibacterial was done by contact bioautography with F254 silica gel stationary phase and mobile phase n-hexane : ethyl acetate (1: 1). The results showed that KHM50 value was 214,98 ���µg/mL while KHM90 at concentration 2943,37 ���µg/mL. Bioautographic results show clear zones on hRf 75-96. Groups of antimicrobial active compounds are probably flavonoids and terpenoids.
