Isolasi, Identifikasi dan Karakterisasi Staphylococcus aureus Asal Bumble Foot dan Artritis pada Ayam Broiler

Staphylococcus aureus pada ayam broiler dapat menyebabkan terjadinya bumble foot dan artritis yang merugikan bagi peternak karena pertumbuhan dan produktivitas ayam terhambat. Penelitian ini bertujuan untuk melakukan isolasi, identifikasi dan karakterisasi Staphylococcus aureus yang diisolasi dari ayam broiler yang mengalami bumble foot dan artritis. Sampel yang digunakan berasal dari peternakan ayam broiler di daerah Kemiri, Sleman, Yogyakarta. Isolasi S. aureus dilakukan dengan menanam spesimen cairan sendi tarsometatarsal dan luka bumble foot ke dalam media Todd Hewitt broth (THB) kemudian diinkubasi pada suhu 370C selama 24 jam. Pertumbuhan koloni bakteri pada plat agar darah (PAD) diamati setelah inkubasi 370C selama 24 jam. Identifikasi terhadap Staphylococcus aureus dilakukan berdasar kemampuan fermentasi pada mannitol salt agar (MSA), pengecatan Gram, uji katalase, uji clumping factor, dan koagulase. Karakterisasi Staphylococcus aureus yang dilakukan meliputi produksi hemolisin, pigmen, dan sifat hidrofobisitas. Dari 27 sampel berhasil diidentifikasi 10 isolat Staphylococcus aureus. Semua isolat bersifat Gram positif, mampu memfermentasikan MSA, positif pada uji katalase, clumping factor dan koagulase. Berdasarkan produksi hemolisin terdapat 8 isolat S. aureus memperlihatkan sifat α hemolitik (80%), 1 isolat β hemolitik (10%) dan 1 isolat memperlihatkan sifat γ hemolitik (10%). Berdasarkan produksi pigmen, 2 isolat S. aureus menghasilkan pigmen berwarna oranye (20%), 6 isolat menghasilkan pigmen berwarna kuning (60%) dan 2 isolat menghasilkan pigmen putih (20%). Semua isolat (100%) bersifat hidrofil. Kata kunci : Staphylococcus aureus, bumble foot, artritis, ayam broiler

Staphylococcus aureus can cause bumble foot and arthritis of broilers which impacted economical loss for farmers due to decrease in growth and productivity of chicken. This research was done to isolate, identificate and characterize of Staphylococcus aureus, isolated from bumble foot and arthritis of chickens. Samples were originated from chicken farm in Kemiri, Sleman, Yogyakarta . The isolation S. aureus was done by planting the specimen from synovial fluid of tarsometatarsal joints and wound of bumble foot lesion in Todd Hawitt Broth (THB) and incubation at 370C for 24 hours . The growth of bacteria on the blood agar plate (PAD) were observed after incubation at 37 0C for 24 hours. Identification of Staphylococcus aureus based on the culture of bacteria on the mannitol salt agar (MSA), Gram staining, catalase, clumping factor, and coagulase tests. Characterization of Staphylococcus aureus were performed production of hemeolysin, pigment and hydrophobicity test. From 27 samples there were 10 isolates positive of Staphylococcus aureus. All isolates were Gram positive, able to ferment MSA, positive for catalase, clumping factor and coagulase test. Based on the production of hemolysin there were 8 isolates of S. aureus showed α-hemolysis (80%), 1 isolate β-hemolysis (10%) and 1 isolate showed γ-hemolysis (10%). Based on the production of pigments there were 2 isolates of S. aureus produced orange pigment (20%), yellow pigment for 6 isolates (60%) and white pigmented for 2 isolates (20%). All isolates (100%) had hydrophilic surfaces. Key words : Staphylococcus aureus, bumble foot, arthritis, broiler
