PENGARUH APLIKASI GEL KITOSAN TULANG CUMI-CUMI (Loligo sp.) 3% TERHADAP ANGIOGENESIS PADA PENYEMBUHAN LUKA GINGIVA (kajian in vivo pada Sprague dawley)

Proses ini didukung dengan keberadaan faktor pertumbuhan, seperti VEGF dan FGF. Kitosan yang terdapat dalam tulang cumi-cumi (Loligo sp.) dapat menstimulasi munculnya faktor-faktor pertumbuhan tersebut. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh aplikasi gel kitosan tulang cumi-cumi (Loligo sp.) 3% terhadap angiogenesis pada proses penyembuhan luka gingiva Sprague dawley. Sebanyak 36 tikus Sprague dawley, jantan, usia 2-3 bulan, berat 200-250 gram dibagi dalam tiga kelompok yaitu kelompok kontrol positif (Aloclair), kelompok perlakuan (Gel kitosan tulang cumi-cumi 3%) dan kelompok kontrol negatif (CMC-Na 2%). Setiap kelompok dibuat perlukaan menggunakan punch biopsy diameter 3 mm, selanjutnya masing-masing kelompok diaplikasikan bahan uji selama 14 hari setiap pagi dan sore. Tikus kemudian dimatikan dengan menginjeksikan overdosis ketamin pada hari ke 3, 5, 7 dan 14 untuk diambil jaringan gingivanya yang selanjutnya digunakan untuk pembuatan preparat histologis dengan pengecatan HE. Pembuluh darah diamati dan dihitung menggunakan mikroskop cahaya dan OptiLab Viewer dalam perbesaran 400 kali. Selanjutnya data yang telah diperoleh dilakukan analisis menggunakan uji ANAVA dua jalur dan Post Hoc metode LSD. Hasil penelitian menunjukkan bahwa terdapat peningkatan jumlah pembuluh darah pada kelompok perlakuan dan kelompok kontrol positif. Angiogenesis pada kelompok perlakuan berjalan lebih cepat dibandingkan pada kelompok kontrol positif. Kesimpulan dari penelitian ini adalah aplikasi gel kitosan tulang cumi-cumi (Loligo sp.) 3% berpengaruh meningkatkan dan mempercepat angiogenesis pada penyembuhan luka gingiva tikus Sprague dawley.
Angiogenesis is an important process in the proliferation phase of wound healing mechanism. This process is supported by the presence of growth factors, such as VEGF and FGF. Chitosan present in the squid bone (Loligo sp.) can stimulate the synthesis of these growth factors. The purpose of this study is to investigate the effect of chitosan gel derived from squid bone (Loligo sp.) 3% on angiogenesis in healing process of Sprague dawley's gingival wound. Thirty six male Sprague dawley rats, 2-3 months of age, weight 200-250 grams were divided into three groups: positive control group (Aloclair), treatment group (3% squid bone chitosan gels) and negative control group (CMC -Na 2%). Each group was injured using punch biopsy diameter of 3 mm, then each group was applied with the testing material for 14 days every morning and afternoon. The rats were then euthanasized by injecting the overdose of ketamine on the 3rd, 5th, 7th and 14th days to extract the gingival tissue used for the preparation of histological samples preparations using HE staining. Blood vessels were observed and calculated using a light microscope and OptiLab Viewer in 400 times magnification. The data were analyzed using two-way ANOVA and Post Hoc LSD method. The results showed that there were increase in number of blood vessels of treatment group and positive control group. The conclusion of this research is the application of squid bone chitosan gel (Loligo sp.) 3% effecting on increasing angiogenesis in gingival wound healing of Sprague dawley rat.
