EFEK ASPIRIN TERHADAP EKSPRESI P-SELEKTIN DAN AGREGASI TROMBOSIT DARAH SUBYEK DISLIPIDEMIA IN VITRO

Latar Belakang : Penggunaan obat anti agregasi trombosit merupakan dasar pengobatan penyakit kardiovaskular. Kelainan komposisi dan fungsi trombosit pada pasien hiperlipidemia menunjukkan bahwa kadar lipid mempengaruhi penanda aktivasi trombosit seperti P-selektin. Tujuan Penelitian : Tujuan penelitian ini adalah untuk mengetahui pengaruh profil lipid terhadap efek aspirin dengan adanya penurunan penghambatan ekspresi pselektin setelah pemberian aspirin dan tanpa pemberian aspirin pada trombosit subyek dispidemia secara in vitro. Serta penurunan penghambatan agregasi trombosit dengan pemberian aspirin dan tanpa pemberian aspirin pada trombosit subyek dislipidemia secara in vitro. Metode : Penelitian memerlukan 5 orang subyek normal lipid dan 5 orang subyek dislipidemia. Setelah diambil darah dari subyek, lalu dibuat PRP selanjutnya dianalisis agregasi trombosit menggunakan Aggram. Serta untuk melihat aktivasi trombosit digunakan penanda p-selektin menggunakan flowsitometri. Hasil Penelitian: Pada kelompok normal lipid, nilai persentase penghambatan agregasi trombosit mengalami peningkatan dibandingkan kelompok dyslipidemia. Pemberian aspirin dengan kondisi dislipidemia secara in vitro menyebabkan penurunan penghambatan agregasi trombosit. Pada kelompok normal lipid menunjukkan 2 subyek mengalami peningkatan persentase penghambatan ekspresi pselectin, dan 3 subyek mengalami penurunan penghambatan ekspresi p-selectin. Sedangkan pada kelompok dislipidemia 3 subyek mengalami peningkatan penghambatan ekspresi p-selectin dan 2 subyek mengalami penurunan persentase penghambatan eksprei p-selectin. Adanya kondisi dislipidemia tidak mempengaruhi penghambatan ekspresi p-selectin oleh aspirin secara in vitro. Kesimpulan : Kondisi dislipidemia dapat mempengaruhi penghambatan agregasi tormbosit dibandingkan subyek normal lipid. Tetapi kondisi dyslipidemia tidak mempengaruhi ekspresi p-selektin dibandingkan normal lipid.

Abstract: The utilization of antiplatelet aggregation is the fundamental of medical treatment for cardiovascular disease. Aberration of composition and platelet function toward patients of hyperlipidemia indicate that level of lipid gives effect to the marker of platelet activation such as P-selectin. Background: The purpose of this research is to determine the influence of profile of lipid toward aspirin effect by the decrease of expression p-selectin inhibition after giving aspirin and without giving aspirin on the platelet to the subject of dispidemia by in vitro. The decrement of thrombocyte aggregation inhibition by giving aspirin and without giving of aspirin to platelet subject of dyslipidemia in vitro. Objective: Research requires 5 persons as normal subjects and 5 persons as dyslipidemia subjects. After the blood sampling of the subject, and make PRP then analyzed the platelet aggregation by using Aggram. As well as to view the platelet activity that used as the p-selectin mark is using flowsitometri. Result: In the normal group of lipids, percentage inhibition of platelet aggregation increased compared to group dyslipidemia. Giving the aspirin in dyslipidemia condition by in vitro leds to decrease in inhibition of platelet aggregation. In normal lipid group, 2 subjects are showing increased percentage of inhibition of the expression of p-selectin, and 3 subjects are decreasing in inhibition of expression of p-selectin. Whereas in the dyslipidemia group, 3 subjects increased the inhibition of the expression of p-selectin and 2 subjects are decreasing in the percentage of inhibition of expression P-selectin. The condition of dyslipidemia does not give effect to the inhibition of p-selectin expression by aspirin in vitro. Conclusion: Dyslipidemia conditions may give affect to the inhibition of platelet aggregation compared to normal subjects lipids. But dyslipidemia condition does not affect the expression of p-selectin compared to normal lipid.
