OPTIMASI EKSTRAKSI BIJI KEDELAI (Glycine max (L.) Merr) TERHADAP KANDUNGAN STIGMASTEROL MENGGUNAKAN RESPONSE SURFACE METHODOLOGY

Kedelai merupakan tumbuhan yang sering dimanfaatkan sebagai bahan pangan, pakan serta bahan baku obat tradisional dan kosmetik. Biji kedelai (Glycine max (L.) Merr) diteliti mengandung berbagai senyawa aktif, salah satunya yaitu stigmasterol. Stigmasterol adalah senyawa sterol yang terdapat dalam tanaman yang memiliki efek antihiperkolesterol. Agar supaya kadar stigmasterol yang didapatkan optimum, diperlukan optimasi kondisi ekstraksi biji kedelai. Penelitian ini dilakukan untuk mengetahui konsentrasi etanol, rasio pelarut-serbuk dan waktu ekstraksi dalam maserasi yang optimum untuk mendapatkan kadar stigmasterol yang tinggi menggunakan Response Surface Methodology. Penelitian dilakukan dua tahap yaitu one-factor-one-time (tahap I) dan Box-Behnken Design (tahap II). Tahap I dilakukan untuk mengetahui faktor yang berpengaruh yaitu konsentrasi etanol (75-95%), rasio pelarut-serbuk (60-140 mL/10 g), dan waktu ekstraksi (1-5 hari). Tahap II dilakukan untuk mendapatkan model persamaan regresi dengan Box-Behnken Design three-level-three-factor berdasarkan software Design Expert 10.01. Model persamaan regresi yang didapat diuji kesesuaian model regresi (lack of fit, sum of square, dan model summary statistics). Hasil penelitian menunjukkan model persamaan regresi yang signifikan yaitu model kuadratik (p<0,05) dengan nilai koefisien determinasi (R2) sebesar 0,9618. Persamaan model orde kedua yang diperoleh yaitu Y=0,011+0,003825X_1+0,0007417X_2-0,000525X_3-0,001483X_1 X_2+0,0005167X_1 X_3-0,001217X_2 X_3+0,0006889X_1^2-0,002144X_2^2 dengan X1: konsentrasi etanol, X2: rasio pelarut-serbuk dan X3: waktu ekstraksi. Kondisi ekstraksi yang optimal yaitu pada konsentrasi etanol 95%, rasio pelarut-serbuk 70,91 mL/10 g, dan waktu 36 jam dengan kadar stigmasterol yang dihasilkan sebesar 0,0163 µg/µL.

Soybean is a plant used as food for human, animal and raw materials of traditional medicine and cosmetics. The soybean seed (Glycine max (L.) Merr) contain various active compounds, stigmasterol is one of them. Stigmasterol is a sterol contained in plants that have antihipercholesterol effects. In order to get optimum stigmasterol level, optimization of soybean extraction condition is required. The aim of this study is to determine the concentration of ethanol, the ratio of solvent-powder, and extraction time to obtain high levels of stigmasterol using Response Surface Methodology. The research was done in two stages: one-factor-one-time (phase I) and Box-Behnken Design (phase II). Phase I to determine the factors that affect in extraction, i.e. the concentration of ethanol (75-95%), the ratio of solvent-powder (60-140 mL / 10 g), and the extraction time (1-5 days). Phase II to obtain regression equation model with Box-Behnken Design three-level-three-factor based on Expert Design software 10.01. The suitability of the regression equation model is tested by lack of fit, sum of square, and summary statistics model tests. The results showed a significant regression model is quadratic model (p <0.05) with the coefficient of determination (R2) of 0.9618. The second-order model equations obtained is Y=0,011+0,003825X_1+0,0007417X_2-0,000525X_3-0,001483X_1 X_2+0,0005167X_1 X_3-0,001217X_2 X_3+0,0006889X_1^2-0,002144X_2^2 with X1: ethanol concentration, X2: solvent-powder ratio and X3: extraction time. The optimal extraction conditions are 95% ethanol concentration, solvent-powder ratio of 70.91 mL / 10 g, and 36 hours extraction with stigmasterol levels generated at 0.0163 ug / mL.
