KLASIFIKASI SUBTIPE PENYAKIT ACUTE MYELOID LEUKEMIA M1, M2 DAN M3 DENGAN SEGMENTASI ACTIVE CONTOUR WITHOUT EDGE DAN MOMENTUM BACKPROPAGATION ARTIFICIAL NEURAL NETWORK

Acute Myeloid Leukemia (AML) merupakan salah satu jenis penyakit kanker yang menyerang sel darah putih jenis myeloid. AML memiliki delapan jenis subtipe, yakni: M0, M1, M2, M3, M4, M5, M6, dan M7. Subtipe AML M1, M2 dan M3 dipengaruhi oleh tipe sel yang sama, myeloblast, sehingga untuk membedakannya diperlukan analisis lebih rinci. Untuk membantu mengatasi kendala tersebut, maka penelitian ini menerapkan bidang studi pengolahan citra digital dan artificial neural network untuk klasifikasi subtipe AML M1, M2 dan M3 berdasarkan tipe sel yang dimiliki. Dalam melakukan klasifikasi subtipe AML M1, M2 dan M3, ada tiga tipe sel yang menjadi faktor pembeda dan perlu diidentifikasi terlebih dahulu, yaitu: myeloblast, promyelosit dan monoblast. Penelitian ini menggunakan algoritma segmentasi active contour without edge (ACWE) untuk segmentasi sel darah putih. Hasil segmentasi kemudian diekstrak fitur sehingga dari masing-masing objek sel didapatkan enam fitur yang diperlukan sebagai parameter pelatihan, yakni: luas area, tepi area atau perimeter, kebundaran, rasio nukleus, mean dan standar deviasi. Proses identifikasi tipe sel memanfaatkan momentum backpropagation dengan berdasarkan kombinasi learning rate, jumlah neuron hidden layer, dan momentum. K-fold cross validation dengan 3 fold digunakan untuk menghindari overfitting pada setiap kelas. Hasil penelitian menunjukkan bahwa algoritma ACWE dapat melakukan segmentasi citra sel darah putih dengan persentase keberhasilan sebesar 83,789% dari total 876 objek sel. Seluruh preparat AML berhasil diidentifikasi sesuai dengan kriteria jumlah tipe sel yang diprediksi melalui pelatihan dengan momentum backpropagation. Kalibrasi pengujian sebanyak lima kali terhadap kombinasi parameter terbaik menghasilkan rata-rata presisi 84,754%, sensitivitas 75,887%, spesifisitas 95,090% dan akurasi 93,569%.

Acute Myeloid Leukemia (AML) is a type of cancer which attacks white blood cells from myeloid. AML has eight subtypes, namely: M0, M1, M2, M3, M4, M5, M6, and M7. AML subtypes M1, M2 and M3 are affected by the same type of cells, myeloblasts, making it needs more detailed analysis to distinguish. To overcome these obstacles, this research is applying digital image processing and artificial neural network for AML subtypes M1, M2 and M3 classification based on the type of the cell. There are three types of cells into differentiating factor within the AML M1, M2 and M3 classification that need to be identified first: myeloblasts, promyelocytes and monoblasts. This research uses active contour without edge (ACWE) algorithms for white blood cells objects segmentation. Image segmentation result is then extracted so that six features required as training parameters from every cells can be obtained, namely: cell area, perimeter, roundness, nucleus ration, mean and standard deviation. Identification of cell types process utilizes momentum backpropagation using combination of learning rate, hidden layer neurons, and momentum. K-fold cross validation with 3 fold is used to avoid overfitting in each class. The result shows that ACWE can be used for segmentation of white blood cells with 83.789% success percentage of 876 total cell objects. The whole AML slides have been identified according to the cell types predicted number through training with momentum backpropagation. Five times testing calibration with the best parameter generates averages of 84.754% precision, 75.887% sensitivity, 95.090% specificity and 93.569% accuracy.
