AKTIVITAS ANTIVIRUS EKSTRAK ETANOLIK DAUN GAHARU Aquilaria malaccensis (Lamk.) dan Gyrinops versteegii (Gilg.) Domke TERHADAP VIRUS DENGUE SEROTIPE-1

Indonesia sebagai negara dengan iklim tropis memiliki resiko kejadian infeksi virus dengue yang tinggi. Virus dengue ditularkan oleh nyamuk Aedes aegypti dan Aedes albopictus, terdiri dari 4 jenis serotipe yaitu DENV- 1, DENV- 2, DENV- 3 dan DENV- 4. Virus dengue serotipe satu merupakan serotipe yang persebarannya paling luas dan sampai saat ini belum ditemukan obat antivirus yang efektif untuk menyembuhkan penyakit ini. Aquilaria malaccensis (AM) dan Gyrinops versteegii (GV) merupakan tanaman gaharu yang saat ini populasinya banyak ditemui di Indonesia. Daun gaharu diketahui memiliki senyawa yang bersifat sebagai antioksidan, antikanker, antibakteri dan imunomodulator. Penelitian mengenai aktivitas antivirus pada ekstrak daun gaharu belum banyak dilakukan. Tujuan penelitian ini adalah untuk mengetahui potensi antivirus ekstrak etanolik AM dan GV terhadap virus DEN-1 dan golongan senyawa ekstrak etanolik daun gaharu terpilih yang berpotensi sebagai antivirus DENV-1. Penelitian ini diawali dengan ekstraksi daun AM dan GV, uji sitotoksisitas ekstrak, propagasi virus dengue serotipe 1, uji antivirus dengan RT-PCR serta pengamatan morfologi sel dan identifikasi senyawa dengan menggunakan KLT. Nilai CC50 ekstrak etanolik AM pada konsentrasi 500 ug/ml, sedangkan ekstrak etanolik GV pada konsentrasi 1000 ug/ml. Uji antivirus menunjukkan bahwa ekstrak etanolik GV memiliki aktivitas antivirus yang lebih baik dengan konsentrasi 1000 ug/ml jika dibandingkan dengan AM dan tidak menyebabkan kematian pada sel vero. Golongan senyawa yang terdeteksi pada ekstrak etanolik GV adalah terpenoid, fenol dan flavonoid.

Indonesia as a country with tropical climate has a high risk of dengue viral infection. Dengue virus is transmitted by Aedes aegypti and Aedes albopictus, and is comprised of four serotypes which are DENV-1, DENV-2, DENV-3 and DENV-4. Dengue virus serotype 1 is the most widespread. Currently, effective antiviral drugs have not been found to treat the four serotypes or even one serotype. Aquilaria malaccensis (AM) and Gyrinops versteegii (GV) are agarwood plants that is mostly found in Indonesia. Several studies proved that agarwood leaves contain secondary metabolites which have pharmacological activities such as antioxidant, anticancer, antibacteria and immunomodulatory. However, further study about antiviral activity of the secondary metabolite in agarwood leaves has not been done. The objectives of this study were to examine the potential antiviral ethanolic extract AM and GV against DENV-1 and identifying which compound of agarwood leaves ethanolic extract that is potential as an antiviral DENV-1. Research was initiated by extraction of AM and GV leaves using ethanol with soxhletation method, extract cytotoxicity assay, propagation of dengue virus serotype 1, examination of antiviral activity by RT-PCR, cell morphology observation, and compound identification using TLC. Results showed that value of CC50 AM ethanolic extracts was at a concentration of 500 ug / ml, whereas GV ethanolic extract was at a concentration of 1000 ug / ml. Antivirus test showed that GV ethanolic extract has better antiviral activity at a concentration of 1000 ug / ml than AM ethanolic extract and did not cause cell death. GV ethanolic extract compounds detected were terpenoids, phenols and flavonoids.
