PENGEMBANGAN KEFIR MENGGUNAKAN ISOLAT MURNI BAKTERI ASAM LAKTAT, BAKTERI ASAM ASETAT, DAN YEAST YANG DIISOLASI DARI KEFIR GEDONO

Kefir adalah minuman susu fermentasi yang diperoleh melalui fermentasi laktat-alkoholik. Sembilan isolat yang diisolasi dari kefir Gedono terdiri dari tiga isolat bakteri asam laktat (L.plantarum GDN-B1, L.plantarum GDN-B2 dan L.plantarum GDN-B3), tiga isolat yeast (C.tropicalis GDN-Y2, C.tropicalis GDN-Y8 dan Yeast GDN-Y7) dan tiga isolat bakteri asam asetat (Acetobacter sp GDN-A2, Acetobacter sp GDN-A3, Acetobacter sp GDN-A5) digunakan untuk pembuatan kefir pada penelitian ini. Tujuan penelitian ini adalah pengembangan inokulum dengan mengkombinasikan isolat murni sebagai starter untuk pembuatan kefir, menentukan starter yang berpotensi menghasilkan granula kefir dan dapat diterima oleh konsumen, mengamati pertumbuhan mikroorganisme serta perubahan pH selama fermentasi pada suhu ruang dan suhu 37oC. Masing-masing isolat (bakteri asam laktat, bakteri asam asetat dan yeast) dikombinasikan sebagai starter untuk pembuatan kefir. Kefir dipersiapkan menggunakan campuran susu UHT dan susu skim bubuk 2% (b/v) yang diinokulasikan dengan starter lalu diinkubasi pada suhu ruang dan suhu 37oC. Uji penerimaan konsumen dilakukan untuk menilai atribut aroma, mouthfeel, kemanisan, keasaman, lingering aftertaste dan secara overall. Kefir yang paling baik berdasarkan uji penerimaan konsumen pada suhu inkubasi yang berbeda diamati pertumbuhan mikroorganismenya dan perubahan pH selama fermentasi. Kefir dengan starter terdiri dari Lactobacillus plantarum GDN-B6, Candida tropicalis GDN-Y2 dan Acetobacter sp GDN-A3 yang diinkubasi pada suhu ruang dan suhu 37oC merupakan kefir yang paling diterima oleh konsumen dengan pH akhir 4,8 dan 5,2 dan diharapkan berpotensi membentuk granula kefir. Kefir is a beverage produced by lactic-alcoholic fermentation of milk using kefir grain. Nine isolates consists of three lactic acid bacteria isolates (L.plantarum GDN-B1, L.plantarum GDN-B2, L.plantarum GDN-B3), three yeast isolates (C.tropicalis GDN-Y2, C.tropicalis GDN-Y8, Yeast GDN-Y7) and three acetic acid bacteria isolates (Acetobacter sp GDN-A2, Acetobacter sp GDN-A3, Acetobacter sp GDN-A5) were isolated from Gedono's kefir and used to produce kefir. The objectives of this research are to develope inoculum by combining pure isolates as starter to produce kefir, to determine starter potential to form kefir grains and aceeptable by consumers, and to study the growth of microorganisms and pH changing during fermentation in ambient temperature and 37oC. Each isolate (lactic acid bacteria, yeasts and acetic acid bacteria) were combined as starter to produce kefir which prepared using mixture of UHT cow milk and skim milk powder 2% (w/v) incubated in ambient and 37oC temperature. Acceptance test to measure aroma, mouthfeel, sweetness, acidity, lingering aftertaste, and overall was performed. The most acceptable kefir from different temperature was studied for the growth of microorganism and pH changing during fermentation. Kefir with starter consist of Lactobacillus plantarum GDN-B6 ,Candida tropicalis GDN-Y2 and Acetobacter sp GDN-A3 incubated at ambient and 37oC temperature was considered as the most acceptable with final pH of 4.8 and 5.2, respectively, and expected potential to form kefir grain. Higher enhancement number of cells and faster fermentation period were occurred at 37oC rather than ambient temperature.
