ABSTRACT Human errors often affect the reliability of diagnostic processes. Automated diagnostic tools can reduce bias from human factor so that the results of the analysis would be more objective, accurate, consistent, and has good repeatability. One of the research and development in this field is the engineering of the microscope system to give the ability to conduct analysis of blood smear samples automatically. An important early stage of blood smear analysis is Good Working Area (GWA) localization on the blood smear sample. This is necessary so that diagnostic processes at later stage, such as the analysis of white blood cells, can work optimally. Automation at this stage is necessary so that the system reliability increases and reduces errors due to selection of working areas which are not ideal. Automation is developed using Digital Image Processing and Artificial Neural Network (ANN). Digital Image Processing is used in feature extraction stage, while the ANN is used as a classifier system with input from image features. Samples acquisition using a 10x agnification objective lens. The input image then divided into 10x10 local areas to provide more detailed input classification. Optimization of the feature significance and the optimization of the ANN parameters is done so that the system can function more efficiently and suitable for low-cost devices. The method have developed successfully to perform classification on the set of local images with 97,6% accuracy. Accumulation of the local images on the frame is used to classify the GWA globally on frame images, and has 96,3% accuracy. The algorithm's processing time to execute classification on each frame image is 6,3 seconds.
