STUDI JENIS DAN POLA DISTRIBUSI GLIKOKONJUGAT PADA ENDOMETRIUM SAPI ACEH (Bos indicus) PADA BERBAGAI STADIUM SIKLUS ESTRUS DENGAN HISTOKIMIA LEKTIN

Kemampuan sapi Aceh bereproduksi di lingkungan yang ekstrim diduga didukung oleh sistem reproduksinya. Penelitian ini dilakukan untuk mempelajari sistem reproduksi sapi Aceh dengan mengidentifikasi jenis dan distribusi glikokonjugat pada endometrium sapi Aceh pada berbagai stadium siklus estrus. Lima belas ekor sapi Aceh betina dewasa pubertas, fase siklus estrus, berat badan 200-300 kg digunakan dalam penelitian ini. Sampel diperoleh dari Rumah Potong Hewan (RPH) Kota Banda Aceh, Provinsi Aceh. Uterus dan ovarium difiksasi dalam larutan Bouin dan diproses secara histologi. Ovarium diwarnai dengan Hematoksilin Eosin (HE) untuk mendeteksi fase siklus estrus, sedangkan uterus diwarnai dengan Alcian Blue-Periodic Acid Shiff (AB-PAS) untuk mendeteksi jenis karbohidrat, dan histokimia lektin (Con-A, WGA, RCA, dan UEA-I) untuk mendeteksi jenis glikokonjugat. Data yang diperoleh disajikan dalam bentuk tabel dan gambar serta dianalisis secara deskriptif. Glikokonjugat D-galaktosa, L-fukosa, D-mannosa/D-glukosa, dan N-asetil-Dglukosamin dapat diidentifikasi di endometrium sapi Aceh pada berbagai stadium siklus estrus. Glikonjugat D- galaktosa sangat kuat teridentifikasi pada korpus dan servik fase proestrus, kornua, korpus dan servik fase estrus, korpus dan servik fase metestrus, serta kornua dan servik fase diestrus. Glikonjugat L-fukosa sangat kuat teridentifikasi pada korpus fase proestrus, korpus dan servik fase estrus dan metestrus serta pada kornua dan servik fase diestrus. Glikonjugat D-manosa/D-glukosa sangat kuat teridentifikasi pada korpus fase proestrus, kornua dan servik fase estrus, korpus fase metestrus serta servik fase diestrus. Sedangkan glikonjugat N-asetil-Dglukosamin sangat kuat teridentifikasi pada korpus fase proestrus dan metestrus. Keberadaan glikokonjugat tersebut diduga berperan penting dalam proses reproduksi yang terjadi di dalam endometrium sapi Aceh pada berbagai stadium siklus estrus.

The reproductive performances of Aceh cattle in extreme environment is supported by its reproductive capacity system. This research was conducted to investigate the glycoconjugate type and its distribution in endometrium of Aceh cattle during estrous cycle. Fifteen Aceh mature cows during estrous cycle, with 200-300 kg of body weight were used in this study. Samples were taken from slaughter house in Banda Aceh Municipality, Aceh Province. Uterus and ovaries samples were fixed in Bouin’s solution, and then histologically process by using parrafin method. Hematoxylin eosin staining on ovaries was performed to identify estrous cycle phase, alcian blueperiodic acid Shiff staining and lectin histochemistry Con-A, WGA, RCA, and UEAI were carried out to detect the carbohydrate type, and the sugar residues of glycoconjugates. The data obtained, then, were presented in the form of tables and figures and they were descriptively analyzed. The glycoconjugates of D-galactose, L-fucose, D-mannose/D- glucose, and Nacetyl- D-glucosamine were detected in the endometrium of Aceh cows at various stage ofestrous cycle. D-galactose glycoconjugate was strongly identified in the corpus and cervix of proestrus stage, in the cornua, corpus, and cervix of estrous stage, in the corpus and cervix of metestrus stage , and in the cornua and cervix of diestrus stage. L-fucose glycoconjugate was strongly identified in the corpus of proestrous stage, in the corpus and cervix of estrous and metestrus stages, and in the cornu and cervix of diestrus stage. D-mannose/D-glucose glycoconjugate was strongly identified in the corpus of proestrus stage, in the cornu and cervix of estrous stage, corpus of metestrus stage, and in the cervix of diestrus stage. While N-acetyle-D-glucosamin glycoconjugate was strongly identified in the corpus of proestrus and metestrus stages. It was suggested that the presence of the glycoconjugate in the endometrium plays important roles on reproduction process of Aceh cow during various stage of estrous cycle.
