PENGARUH PENCUCIAN DAN SUHU PENYIMPANAN TERHADAP KUALITAS SPERMA KAMBING PERANAKAN ETTAWA YANG DIENCERKAN DENGAN GLUKOSA SITRAT KUNING TELUR

Tujuan dari penelitiao ini axialah untuk ntengetahui pengaruh pencucian sperma kambing Peranakun Bltawa (PE) yang diencerkan mernakai pengencer glukosa sitrat kuning telur 15% pada pendingiwaa dan pembekuan terhadap kualitas sperma yang meliputi motilitas, pH dan spermatozoa hidup. Sperma dari dua ekor kambing PE jantan ditampung dengan metode vagina buatan seminggu sekali seiama 1 bulan, kemudian setengah bagian sperma dicuci dengan Krebs ringer phosphat, Sperma yang dicuci dao tidak dicuci selaojutnya didinguikan atau dibekukan, seiama 24 dan 48 jam. Penelitian ini dianalisis dengan anaiisis variansi atau kovariansi dalam rancangan a.cak lengkap pola faktorial 2x2x3 dan diianjutkan dengan uji Duncan’s multiple range test dan West. Hasil penelitian menunjukkan perbedaan yang sangat nyata pada perlakuan pencucian, temperatur penyimpanan clan waktu penyimpanan terhadap motilitas speraiatozoa (P<0,01 ) Terdapat interaksi yang nyata (P<0,05) dan sangat nyata, (PW),01) antara pencucian sperma dengan waktu penyimpanan dan antara temperatur dengan waktu penyimpanan terhadap motilitas spermatozoa Hasil penelitian tnenunjukan perbedaan yang sangat: nyata pada perlakuan pencucian dan waktu penyimpanan terhadap pH sperma (P<0,01). Terdapat interaksi yang sangat nyata antara pencucian dengan waktu penyimpanan, antara temperate dengan waktu penyimpanan dan antara ketiga perlakuan terhadap pH sperma (P<0,01). Terdapat perbedaan yang nyata (P<0,05) clan sangat nyata (P<0,01.) pada perlakuan pencucian dan waktu penyimpanan terhadap persentase speraiatozoa hidup. Tidak terdapat interaksi yang nyata antara ke tiga perlakuan terhadap persentase spermatozoa hidup (PM),05), Dan basil yang diperoleh dapat disimpuikan bahwa Pencucian sperma dapat meaipertahankan motilitas speraiatozoa serta memperiambat penurunan pH sperma pada saat didiriginkan dan dibekukan. Selanjutnya untuk keperluan inseminasi buataii (18) diary intern menggunakan sperma kambing Peranakan Ettawa yang tel.ah dicuci.. Sperma. kambing Peranakan Ettawa yang dicuci dao dibekukan tolah mmxivmlu syarai imiuk inseiainasi buatan, karena motilitasnya telah mencapm 40%. lliis study was carried out to evaluate the effect of seminal plasma removal before diluted with 15% glucose egg yolk citrate extender on the quality of frozen and cold Ettawa grade buck semen, including motility, acidity (pH) and percentage of living spermatozoa, Semen from two Ettawa grade buck semen were collected using artificial vagina method, weekly for four months. The neat semen was then splitted into two halves. A half of the neat semen was removed by washing it with Krebs ringer phosphat, then both of them were extended using 15% glucose egg yolk citrate, and then the neat semen was cooled and frozen for 24 and 48 hours. Data were analyzed using variance or covariance analysis in 2x2x3 factorial design. Then it was continued with Duncan's multiple range test. Result of the study showed differences (P<.01) in washing treatment, preservation temperature and time of preservation on the motility of spermatozoa Interaction (P<.01) were found between preservation temperature and time of preservation. There was interaction (P<.05) between washing treatment and preservation temperature on the motility of spermatozoa Result of the study showed differences (P<.01) between washing treatment and time of preservation on the semen acidity. There were interaction (P<.01) between washing treatment and preservation temperature and between preservation temperature and time of preservation on the semen acidity. The differences (P<.01 and P<.05) were found in temperature and washing treatment on the percentage of life spermatozoa, but no significant interaction were found among all of the treatments on the percentage of life spermatozoa From the result of the study it can be concluded that semen washing can keep the motility of spermatozoa and hold on decreased of semen acidity when being cooled and frozen. For artificial Insemination purpose, using washed semen were better. Washed-frozen Ettawa grade buck semen in the study were ready for use, because the motility was up to 40%.
